Abstract Bone is constantly being remodeled, and this process orchestrated by a dynamic crosstalk of bone cells, including osteoclasts, osteoblasts, osteocytes. Recent evidence suggests that cellular metabolism plays crucial role in the differentiation function cells facilitates adaptation to changes microenvironment. Moreover, affects whole-body energy metabolism. However, it not yet completely understood how different coordinate metabolic processes under physiological conditions, altered contribute pathological conditions where balance among disrupted. Therefore, gaining better understanding distinct requirements can provide insights into dysfunction be used identify new therapeutic approaches treat diseases. Here, we discuss recent advances reprogramming cells.

"Nano", which derives from the Latin word nanus and means dwarf, refers to a very small unit of measurement equal one billionth meter. Nanotechnology, deals with manipulation matter at atomic molecular level, has an application area in animal husbandry as well many fields. Nano-sized feed additives, have come forefront livestock sector recent years, become innovative used increase nutritional value feeds optimize health performance. Since these additives are nano-sized particles increased specific surface area, they can positive effect on number factors such digestibility, nutrient absorption, immune system, growth development. Minerals form nanoparticles bioavailability by passing through intestinal wall body cells faster compared larger particles. The nano level substance not only increases productivity animals, but also brings potential improve functionality molecules. Nano digestion absorption feed, allowing animals benefit more effectively. However, there several challenges associated this approach. These include for endotoxin production, reduced due interaction natural nutrients, possibility nanoparticle accumulation body, risks, ethical considerations, environmental concerns some negative effects interference nutrients that be avoided encapsulation. Hemoglobinopathies are monogenic disorders affecting hemoglobin synthesis. Thalassemia and sickle cell disease (SCD) considered the two major hemoglobinopathies. is a genetic disorder one of hemoglobinopathies determined by an impairment globin chain production, which causes alteration erythropoiesis, improvement in hemolysis, iron homoeostasis. In SCD, mutations on β-globin results substitution glutamic acid valine with consequent formation Hemoglobin S (HbS). Several factors involved bone metabolism patients hemoglobinopathies, among them hormonal deficiency, marrow hyperplasia, overload, inflammation, increased turnover. Bone result balance maintenance between deposition resorption, osteoblasts (OBs) osteoclasts (OCs). An this responsible for onset diseases, such as osteoporosis (OP). Therefore, here we will discuss possible therapeutic strategies to contain and/or counteract health these patients, taking into consideration not only pharmacological treatments already used clinical armamentarium, but also new strategies.

The relationship between lipid metabolism and bone mineral density (BMD) is still not fully elucidated. Despite the presence of investigations using osteoporotic animal models, clinical studies in humans are limited. In this work, untargeted lipidomics profiling liquid chromatography-mass spectrometry (LC-MS) analysis human serum samples was performed to identify profile associated with low (LBMD), a subsequent examination potential biomarkers related OP risk prediction or progression. A total 69 participants were recruited for cohort study, including group (OP, n = 25), osteopenia (ON, 22), control (Ctrl, 22). LBMD included ON patients. effect confounding factors such as age, gender, profile, body mass index (BMD), chronic diseases, medications excluded from dataset. results showed clear separation clustering Ctrl (Q2 0.944, R2 0.991), indicating significant difference lipids profile. addition, 322 putatively identified molecules dysregulated, 163 up- 159 down-regulated LBMD, compared Ctrl. most significantly dysregulated subclasses phosphatidylcholines (PC) (n 81, 25.16% all 322), followed by triacylglycerol (TG) 65, 20.19%), then phosphatidylethanolamine (PE) 40, 12.42%). groups glycerophospholipids, LPC (7.45%), LPE (5.59%), PI (2.48%) also LBMD. However, other lipids cholesterol have also been found to play critical role homeostasis can contribute overall bioenergetic capacity osteoblasts. In addition, several epidemiological studies link between elevated cholesterol, cardiovascular disease, enhanced risk osteoporosis, increased metastasis cancer patients. This review focuses on how its derivatives, cholesterol-lowering medications (statins) regulate osteoblast function formation. It highlights molecular mechanisms underlying cholesterol–osteoblast crosstalk.

We conducted this cross-sectional study to investigate the independent associations between lipid metabolites and osteoporotic fractures among participants aged 40–69 years from UK Biobank. Serum lipid, lipoprotein levels nuclear magnetic resonance (NMR) based metabolic biomarkers were measured at baseline. multivariable logistic analyses potential concentrations of in both men women. The odds ratios (ORs) for calculated on their lowest tertile. Over a median follow-up period 15 years, total 978 4515 women diagnosed with osteoporosis, whereas 138 327 encountered incident fractures. Statistically significant disparities identified NMR-based compared those without. Out 144 distinct known, 35 exhibited patients osteoporosis. Following adjustment confounding factors, degree unsaturation (p = 0.0066) docosahexaenoic acids 0.0011) male increased risk And high level different HDL 0.0153), 3-Hydroxybutyrate 0.0012) Sphingomyelins 0.0036) decreased female patients. This outcome indicates that these may potentially have unique roles independently contributing occurrence
