This study aimed to identify risk factors and potential targets associated with non-responsiveness 5-Aminosalicylic acid (5-ASA) in patients ulcerative colitis (UC). Patients mild moderate UC were grouped based on their responsiveness 5-ASA. Regression analysis was conducted for non-responsiveness, while Mendelian randomization (MR) used evaluate the targets. Among 50 (11 non-responders 39 responders), exhibited higher incidences of fever, back pain, allergy history, ankylosing spondylitis, as well elevated monocyte count alanine aminotransferase (ALT) level, but a lower aspartate aminotransferase-to-ALT (AST/ALT) ratio (all P < 0.05). Allergy history factor (OR 140.50, = 0.021), high AST/ALT protective 0.001, 0.023) non-responsiveness. The area under receiver operating characteristic curve 0.80. MR showed that inhibition inhibitor nuclear kappa-B kinase subunit alpha (CHUK) by 5-ASA, targeting ALT, increased 1.02, 0.002). Our findings suggested CHUK identified target ALT-related Trial Registration: ChiCTR2400084086|| http://www.chictr.org.cn/ , Chinese Clinical Registry, 2024-05-10.

Estrogen receptors (ERs) play a critical role in breast cancer (BC) development and progression, with ERα being oncogenic ERβ exhibiting tumor-suppressive properties. The interaction between ER signaling other molecular pathways, such as PI3K/AKT/mTOR, influences tumor growth endocrine resistance. Emerging research highlights the of prebiotics modulating gut microbiota, which may influence estrogen metabolism, immune function, therapeutic responses BC. This review explores impact on receptor modulation, microbiota composition, regulation, metabolic pathways cancer. potential adjunctive therapies to enhance treatment efficacy mitigate chemotherapy-related side effects is discussed. A comprehensive analysis recent preclinical clinical studies was conducted, examining reprogramming short-chain fatty acids (SCFAs) derived from prebiotic fermentation epigenetic regulation resistance also evaluated. Prebiotics were found modulate microbiota-estrogen axis, reduce inflammation, responses. SCFAs demonstrated selective downregulation reprogramming, suppressing growth. Synbiotic interventions effects, improving quality life patients. offer promising avenue for prevention therapy by pathways. Future trials are needed validate their treatments management.

Background Rosacea is a common chronic inflammatory skin disease. Limited studies reported the association between plasma lipidome and rosacea. Methods We employed two-sample Mendelian randomization (MR) study to assess causality Plasma genome-wide (GWAS) data were collected. The inverse variance weighted (IVW) method was utilized as principal in our study; we also used MR-Egger, median, simple mode, mode methods. MR-Egger intercept test, Cochran’s Q MR-Pleiotropy RESidual Sum Outlier (MR-PRESSO), leave-one-out analysis conducted identify heterogeneity pleiotropy. Results A total of 179 lipid species analyzed; among them, five closely related Two sterol ester [sterol (27:1/22:6) (27:1/15:0)], two phosphatidylethanolamine [phosphatidylethanolamine (O-18:2_20:4) (18:0_20:4)], one sphingomyelin [sphingomyelin (d34:0)] causally associated with rosacea ( P < 0.05). All them play protective roles patients No or pleiotropy observed. Conclusion This provided new evidence relationship Our MR suggested that progression. These could be novel effective ways treat

The causative relationship between lung cancer subtypes and lipids is yet unknown. This research aims to elucidate the potential link connecting lipid levels subtypes, particularly focusing on non-small cell (NSCLC), using Mendelian randomization (MR) method meta-analysis. Summary statistics were obtained from genome-wide association study (GWAS) datasets. A comprehensive MR analysis was performed explore causal role of in NSCLC subtypes. To ensure reliability results, an external dataset used for validation, a meta-analysis further synthesis. Two 179 examined showed connection with adenocarcinoma (LUAD) three squamous carcinoma (LUSC). Specifically, phosphatidylcholine (PC) (16:0_20:4) PC (18:0_20:4) might be connected elevated risk LUSC, but (18:0_20:2) linked decreased risk. It discovered that (16:1_20:4) raise LUAD. Complex metabolic pathways, especially involving PC, are present NSCLC, distinct isomers may influence various molecular different ways.

IgA nephropathy (IgAN) is a leading cause of chronic kidney disease, often associated with dyslipidemia and immune dysfunction. This study employs Mendelian randomization (MR) to investigate the causal relationship between plasma lipidomes IgAN, focus on potential mediating role cells. We analyzed 179 genetically predicted IgAN gene using two-sample (TSMR) multivariable MR based summary-level data from genome-wide association study, results were validated by liquid chromatography-mass spectrometry. Furthermore, we quantified proportional effect cell-mediated TSMR. identified significant relationships 3 risk examining lipidome traits as exposures. To whether impact lipid groups specific, performed TSMR analyses exposure factors 4 nephritides outcomes. Specifically, only phosphatidylinositol (18:1_20:4) was found have negative incidence (IVW method, p = 0.01, OR 0.71, 95% CI 0.55 - 0.92). Our further analysis focused 8 cells IgAN. 2 cell phenotypes that may contribute (18:1_20:4)-mediated careful screening. findings provide robust genetic evidence supporting link acting mediators. Phosphatidylinositol emerges promising biomarker for stratification, early detection, therapeutic intervention. Modulating its levels offer novel avenues management.
