Abstract Inflammation involving adipose macrophages is an important inducer of obesity. Regulating polarization and improving the inflammatory microenvironment tissue a new strategy for treatment An amphiphilic chondroitin sulfate phenylborate derivative (CS-PBE) was obtained by modifying main chain with hydrophobic small molecule phenylborate. Using CS-PBE self-assembly, macrophage targeting, reactive oxygen species (ROS) release celastrol (CLT) encapsulation were achieved. The cytotoxicity, cellular uptake, internalization pathways transmembrane transport efficiency micelles studied in Caco-2 RAW264.7 cells. Hemolysis organotoxicity tests performed to assess safety platform, while its therapeutic efficacy investigated high-fat diet-induced obese mice. Multifunctional targeting ROS clearance capabilities developed improve CLT treating In vitro studies indicated that had better ability target M1 macrophages, protective effects on mitochondrial function, reduce number LPS-stimulated M2 scavenge macrophages. vivo have shown inflammation significantly toxicity summary, could alleviate inflammation, suggesting are highly promising approach
