Abstract The scarcity of clinically available antifungal drugs, coupled with frequent side effects and the accelerating emergence fungal drug resistance, underscores pressing demand for novel drugs. Host defense peptide (HDP) assemblies are one ideal alternatives to drugs due distinctive advantages nanostructures, but still face challenges in terms vivo stability, biosafety, facile synthesis. Poly‐β‐peptides a class polypeptide mimics that possess excellent biocompatibility resistance proteolysis, however, their antimicrobial have yet be explored. Herein, poly‐β‐peptide poly(sarcosine) integrated design self‐assembled functionalized HDP mimics, while establishing one‐pot, open‐vessel synthesis method block‐like poly‐β‐peptide. synthesized multifunctional exhibit potent highly selective activity against drug‐resistant fungi effectively eradicate mature biofilms, demonstrate efficacy. Notably, insusceptible acquire owing mechanisms associated organelle destruction reactive oxygen species. These results potential strategy developing combating infections.
