During the process of metastasis, cancer cells transmigrate through endothelial barrier vascular walls. Cancer interact with cells, leading to changes their mechanical properties. While this interaction has been extensively studied from perspective (CCs), properties endothelium remain underexplored, even though they may play a crucial role in regulating process. In study, we used microrheology AFM-based approach demonstrate that (ECs) are altered when cultured either CC-conditioned medium or direct contact cells. We found viscoelasticity ECs decreased diluted medium. Furthermore, cell at short times (less than 30 min) also showed significantly lower stiffness and viscoelastic moduli. parallel, observed reduction quantity actin fibers clusters within EC cytoskeleton, resulting less structured network facilitate CC transmigration. Overall, our findings indicated rheological can be directly physical CCs indirectly via CC-secreted substances. This suggests actively responds presence rather serving only as passive barrier.
