Abstract Background Chronic prostatitis (CP)/chronic pelvic pain syndrome is the most common urological disorder in young and middle-aged men. A previous study showed that melatonin attenuates prostate inflammation through Sirt1-dependent suppression of nonobese diabetic–like receptor thermal protein domain-associated 3 inflammasome mouse models experimental autoimmune (EAP). However, main differentially expressed proteins (DEPs) melatonin-treated mice with EAP have not yet been fully identified. Materials Methods Mouse were established. The pathological morphology tissues was observed using hematoxylin-eosin staining. sensitivity assessed suprapubic allodynia. Inflammation-related cytokines detected an enzyme-linked immunosorbent assay. These methods used to validate successful establishment model. Tandem mass tag proteomics identify proteomic markers mice. Next, we visualized DEPs bioinformatic analyses. Finally, measured expression mitochondrial creatine kinase 1 gap junction β-1, which identified by tandem all groups, Western blotting explore key involved anti-inflammatory effects on EAP. Results We 5910 proteins, quantitative information available for over 85% total. found 53 between control groups 22 EAP-Melatonin (Mel) groups. Bioinformatic analysis suggested significant alterations immunosuppression, inflammatory chemotaxis, energy metabolism signaling treated melatonin. confirmed blotting. Conclusions Melatonin effectively relieves CP/chronic syndrome-related symptoms Mitochondrial kinases are potential treatment melatonin, these biomarkers may provide direction studying molecular mechanisms CP.
