To investigate the effects and underlying mechanisms of indirubin in treating ALL using network pharmacology experimental validation. Potential targets indirubin- ALL-related genes were identified public databases. Core filtered through protein-protein interaction analysis Cytoscape. Gene Ontology (GO) Kyoto Encyclopedia Genes Genomes (KEGG) pathway enrichment analyses conducted to explore potential against ALL. Drug-disease-functional annotation-signaling maps constructed. Molecular docking between core proteins was performed AutoDock Vina software. Finally, both vitro vivo experiments validate these findings. PPI eight ALL: AKT1, CASP3, mammalian target rapamycin. GO KEGG suggested that mechanism action involves multiple biological functions signaling pathways, with PI3K-AKT likely playing a central role. findings further confirmed strong binding affinity for targets. Both demonstrated inhibited cell proliferation induced cycle arrest apoptosis; may involve pathway. The increasing prevalence of environmental pollutants has raised public concern about their potential role in diseases such as atherosclerosis (AS). Existing studies suggest that chemicals, including bisphenol S (BPS), may adversely affect cardiovascular health, but the specific mechanisms remain unclear. This study aims to elucidate effects BPS on AS and underlying mechanisms. Through an extensive search databases ChEMBL, STITCH, SwissTargetPrediction, SuperPred, SEA, GEO, we identified 34 targets related BPS-induced AS. A target network was constructed using STRING platform Cytoscape software. GO KEGG functional enrichment analysis DAVID database revealed promote occurrence by interfering with critical biological processes glutathione metabolism, nitrogen tyrosine metabolism. followed selection 4 core targets-aminopeptidase n (ANPEP), alcohol dehydrogenase 5 (ADH5), lysosomal pro-x carboxypeptidase (PRCP), microsomal s-transferase 1 (MGST1)-using five machine learning methods. These play a pivotal Furthermore, molecular docking confirmed tight binding between these targets. In conclusion, this provides theoretical framework for understanding contributes scientific evidence development prevention treatment strategies triggered exposure.

Abstract: Metabolic dysfunction-associated steatotic liver disease (MASLD), previously known as non-alcoholic fatty (NAFLD), is becoming more prominent globally due to an increase in the prevalence of obesity, dyslipidemia, and type 2 diabetes. A great deal studies have proposed potential treatments for MASLD, with few them demonstrating promising results. The aim this study was investigate effects (-)-epicatechin (EPI) on development MASLD vitro model using HepG2 cell line by determining metabolic viability cells levels PPARα, PPARγ, GSH. were pretreated 10, 30, 50, 100 μM EPI 4 h assess lipid metabolism. culture established hepatocytes which exposed 1.5 mM oleic acid (OA) 24 h. Moreover, colorimetric MTS assay used order determine cells, PPARα PPARγ protein determined enzyme-linked immunosorbent (ELISA), accumulation visualized Oil Red O Staining method. Also, intracellular glutathione (GSH) measured level oxidative stress. shown treated OA. after incubation OA, significantly decreased, a 71%, compared EPI, where 74–86% respect concentration experiment. Furthermore, GSH exhibited decrease response increasing concentrations. Pretreatment has demonstrated effect vitro. Therefore, considering that mediates metabolism it should be considered hepatoprotective agent future research.
