Bisphenol A (BPA) is a prevalent environmental contaminant found in plastics and known for its endocrine-disrupting properties, posing risks to both human health the environment. Despite widespread presence, impact of BPA on papillary thyroid cancer (PTC) progression, especially under realistic conditions, not well understood. This study examined effects PTC using 3D tumor spheroid model, which better mimicked complex interactions within tissues compared traditional 2D models. Our findings demonstrated that BPA, at environmentally relevant concentrations, could induce significant changes cells, including decrease E-cadherin expression, an increase vimentin reduced thyroglobulin (TG) secretion. These suggest exposure may promote epithelial–mesenchymal transition (EMT), enhance invasiveness, reduce cell differentiation, potentially complicating treatment, by increasing resistance radioiodine therapy. research highlights BPA’s hazardous nature as emphasizes need advanced vitro models, like spheroids, assess posed such chemicals. Thyroid cancer is the most common malignant tumor of endocrine system, and evidence suggests that post-translational modifications (PTMs) epigenetic alterations play an important role in its development. Recently, there has been increasing linking dysregulation ubiquitinating enzymes deubiquitinases with thyroid cancer. This review aims to summarize our current understanding ubiquitination-modifying cancer, including their regulation oncogenic pathways proteins.
