Since late 2021, Omicron variants have dominated the epidemiological scenario as most successful severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2) sublineages, driving new and breakthrough infections globally over past two years. In this study, we investigated for first time host salivary response of COVID-19 patients infected with (BA.1, BA.2, BA.4/5) by using an untargeted four-dimensional data-independent acquisition (4D-DIA)-based proteomics approach. We identified 137 proteins whose abundance levels differed between positive negative groups. Salivary signatures were mainly enriched in ribosomal proteins, linked to mRNAviral translation, protein synthesis processing, immune innate, antiapoptotic signaling. The higher 14-3-3 (YWHAG, YWHAQ, YWHAE, SFN) saliva, reported here, may be associated increased infectivity improved viral replicative fitness. also seven (ACTN1, H2AC2, GSN, NDKA, CD109, GGH, PCYOX) that yielded comprehension into infection performed outstandingly screening a hospital setting. This panel presented enhanced anti-COVID-19 anti-inflammatory signature, providing insights disease severity, supported comparisons other proteome data sets. signature provided valuable host's SARS-CoV-2 infection, shedding light on pathophysiology COVID-19, particularly cases mild disease. It underscores potential clinical applications saliva settings. Data are available via ProteomeXchange identifier PXD054133.

COVID-19 has been a challenge at the healthcare level not only in early stages of pandemic, but also subsequent appearance long-term COVID-19. Several investigations have attempted to identify proteomic biomarkers an attempt improve clinical care, guide treatment and predict possible patient outcomes. Proteins such as C-reactive protein (CRP) or interleukin 6 (IL-6) are clear markers severe disease, many others proposed that could help risk stratification prediction specific complications. This review aims bring together most relevant studies this regard, providing information notable relation found date.

Approximately 10-30% of individuals continue to experience symptoms classified as post-acute sequelae coronavirus disease 2019 (COVID-19 (PASC)). PASC is a multisystem condition primarily characterized by respiratory symptoms, such reduced diffusing capacity for carbon monoxide (DLco). Although many studies have investigated the pathogenesis acute COVID-19, long-term molecular changes in COVID-19 convalescents with remain poorly understood. We prospectively recruited 70 who had been diagnosed from 7 January 2020 29 May (i.e., convalescents); we performed follow-up visits at 6 months, 1 year, 2 years, and 3 years after hospital discharge. Thirty-five healthy controls (CONs), physical examination center before pandemic, served comparison group. explored proteomic metabolomic profiles 174 plasma samples 35 CONs. comprehensive analysis investigate host up Our multi-omics revealed activation cytoskeletal organization glycolysis/gluconeogenesis, well suppression gas transport adaptive immune responses, convalescents. Additionally, metabolites involved glutathione metabolism; alanine, aspartate, glutamate ascorbate aldarate metabolism were significantly upregulated Pulmonary abnormalities persisted convalescents; impaired (DLco) was most prominent feature. used this profile develop model involving one protein (heterogeneous nuclear ribonucleoprotein K (HNRNPK)) two (arachidonoyl-EA 1-O-(2r-hydroxy-pentadecyl)-sn-glycerol)) identification abnormal DLco. Lipid rafts are specialized microdomains within cellular membranes enriched with cholesterol and sphingolipids that play key roles in organization, signaling, homeostasis. This review highlights their involvement protein clustering, energy metabolism, oxidative stress responses, inflammation, autophagy, apoptosis. These findings clarify influence on trafficking, adhesion while interacting the extracellular matrix, cytoskeleton, ion channels, making them pivotal progression of various diseases. further addresses contributions to immune including autoimmune diseases, chronic cytokine storms. Additionally, role as entry points for pathogens has been demonstrated, raft-associated receptors being exploited by viruses bacteria increase infectivity evade defenses. Disruptions lipid raft dynamics linked signaling defects, which contribute metabolic, neurodegenerative, cardiovascular underscores potential therapeutic targets, discussing innovations such engineered transplantation. Advances analytical techniques mass spectrometry have expanded our understanding composition dynamics, opening new directions research. By consolidating current insights, we highlight need exploration molecular mechanisms.
