Background: Depression is one of the most serious and common health problems among youth population responsible for initiation many diseases. As per World Health Organization, 3.8% suffers from mental depression, globally. The monoamine oxidase-A (MAO-A) enzyme degradation neurotransmitters leading to lower levels neurotransmitters. Methods: Chalcones (C1-C15) were synthesized by reacting substituted acetophenone with various benzaldehydes in a basic ethanolic solvent at 80 °C under microwave irradiation conditions. To compare reaction time product yield, conventional method synthesis chalcones was also performed. spectroscopically characterized screened initially inhibitory activities against MAO-A MAO-B. best active compounds undertaken IC50 determination followed reversibility inhibition analysis antioxidant assay. Moreover, silico molecular docking ADME pharmacokinetic investigations accomplished. Results: Most inhibited MAO-A, specifically, C14 C6 exhibited highest values 7.91 ± 0.08 μM 8.45 0.19 μM, respectively. Both these reversible displaying up 60% recovery enzymatic activity when diluted substrate (Tyramine). results study indicated scores −9.56 Kcal/mol (C14) −9.45 (C6) π-π stacking interaction crucial amino acid Trp-397. determined cross blood–brain barrier (BBB) displayed favorable gastrointestinal (GI) absorption. Further, assay demonstrated that possess modest free radical scavenging potential. Conclusions: This potential morpholine-substituted as promising pharmacophore development novel antidepressant lead compounds.

Abstract Objective: Sleep problems are frequent and impairing in adolescents with ADHD. This randomized controlled trial investigates the effectiveness of a newly developed CBT sleep intervention for ADHD – SIESTA. Method: Adolescents (N = 92, Mage 14.36, SD 1.43, 47% males) were to receive SIESTA next treatment as usual targeting (SIESTA+TAU) or TAU only. Adolescent parent ratings, actigraphy diaries, collected at pre-, post-, 4 month follow-up test. A linear mixed effects model was used an intent-to-treat approach. Results: Results indicated significant improvement SIESTA+TAU compared from pre- post-test on hygiene (partial eta squared .21), chronic reduction .15), sleep-wake problem behaviors .05). Actigraphy diaries showed no differences, both groups improving diaries. The improvements maintained .09). Of secondary outcomes, depressive symptoms reduced significantly more than only Conclusions: study indicates that is effective hygiene, perceived problems, However, maintain long-term effects, booster sessions may be beneficial.
