previous
editions,
the
fifteenth
annual
workshop
of
European
Network
for
Breast
Development
and
Cancer
(ENBDC)
on
Methods
in
Mammary
Gland
Biology
was
held
from
2nd
to
4th
May
Weggis,
Switzerland.
Over
course
this
meeting,
participants
followed
discussed
presentations
a
roster
internationally
renowned
invited
speakers
selected
abstracts,
complemented
with
two
poster
sessions
covering
exciting
unpublished
results.
The
covered
projects
normal
mammary
gland
development,
breast
cancer
evolution
metastasis,
as
well
epigenetic
metabolic
regulation
cancer.
As
last
year,
early
career
researchers
(ECR)
hosted
pre-workshop
day,
when
stage
provided
allow
PhD
students
postdocs
showcase
their
results,
they
had
ample
time
discussions
experimental
settings
planning,
while
interacting
several
guests.

ABSTRACT
Basement
membrane
(BM)
supports
and
regulates
the
structural
integrity,
function
differentiation
of
epithelial
tissues
protects
against
stromal
invasion
breast
cancer
cells.
Here
we
show
that
LAMA5,
a
central
BM
component,
is
critical
for
luminal
mammary
tumour
initiation,
controls
tumor
progression
phenotype
development.
LAMA5
overexpressed
in
both
human
mouse
tumours
downregulation
attenuates
growth
Prepubertal
deletion
Lama5
MMTV-PyMT
mice
results
marked
reduction
emergence
early
hyperplasias,
along
with
shift
from
mature
hormone
receptor
(HR+)
positive
towards
HR+
progenitor
phenotype,
widespread
alterations
extracellular
matrix
Fibroblast
factor
(Fgf)
signalling
genes,
including
overexpression
Fgf
2
(Fgfr2)
Single
allele
deletion,
but
not
biallelic
additionally
inhibits
advanced
carcinomas.
However,
inhibition
receptors
restores
decrease
induces
apoptosis
biallelic-
Lama5-
deleted
organoids
without
affecting
wildtype
organoids.
Our
analyses
demonstrate
role
component
initiation
reveal
mechanisms
matrix-epithelial
interplay
maintenance.
