Abstract Exosomes are a subpopulation of nanosized extracellular vesicles (EVs), formed by lipid bilayer and naturally secreted cells. They transport RNA, microRNAs, bioactive proteins, lipids play an important role in intercellular communication. promising alternative cell-free therapy regenerative medicine, immunotherapy, drug delivery. The implementation new exosomes treatments requires knowledge its concentration, purity, full characterization. However, comparing different studies is highly challenging due to the lack validated methodologies for isolation determination, as well well-characterized reference standards. In this work, human uterine cervical mesenchymal stem cell (hUCESC) EVs have been isolated ultracentrifugation characterized, discussing current limitations characterization methods. First, total protein assays heavily influenced free-protein contaminations which confirm need employing several methods vesicular determination. This purity variation seems origin more complex mediums originate matrix interferences. Size exclusion high performance liquid chromatography has demonstrated methodology assessment hUCESC-EVs commercial (adipose cells serum). results found low highlighting that must be included EVs. Abstract Malignant melanoma represents a particularly aggressive type of skin cancer, originating from the pathological transformation melanocytes. While conventional interventions such as surgical resection, chemotherapy, and radiation therapy are available, their non-specificity collateral damage to normal cells has shifted focus towards immunotherapy notable approach. Extracellular vesicles (EVs) naturally occurring transporters, capable delivering tumor-specific antigens directly engaging in immune response. Multiple types EVs have emerged promising platforms for vaccination. The effectiveness EV-based vaccines manifests ability potentiate response, by activating dendritic (DCs) CD8+ T lymphocytes, through engineering synergy antigen presentation targeted delivery. Here, this review mainly focuses on construction strategies EV various sources, effects, immunological mechanisms treating melanoma, well shortcomings future perspectives field. These findings will provide novel insights into innovative exploitation therapy.
