The importance of gut-liver axis in the pathophysiology metabolic dysfunction-associated fatty liver disease (MAFLD) is being investigated more closely recent times. Increased pathogenic taxa like Desulfovibrionaceae, Peptostreptococcus, Clostridium, Terrisporobacter noted. not only reversed this but also modulated taxonomic signatures beneficial microbes Ruminococcus, Lactobacillus, Alloprevotella. Therefore, potential restore eubiosis could be utilized as an intervention strategy prevention MAFLD related disorders.

Punicalagin (PUN) was isolated from the peel of pomegranate (Punica granatum L.), is a polyphenol with anti-inflammatory, hepatoprotective, and antioxidant activities. However, it remains unclear whether PUN alleviates inflammation anti-inflammatory mechanisms in pro-inflammatory cytokines-induced human keratinocyte HaCaT cells. Here, we investigated that tumor necrosis factor-alpha (TNF-α) interferon-gamma (IFN-γ) mixture-stimulated cells were treated various concentrations PUN, followed by analyzed expression inflammation-related mediators evaluate anti-inflammatory-related pathways. Our results demonstrated ≤ 100 μM did not reduce cell viability, ≥ 3 sufficient to decrease interleukin-6 (IL-6), IL-8, monocyte chemoattractant protein-1 (MCP-1), chemokine ligand 5 (CCL5), CCL17 CCL20 concentrations. Ethnopharmacological use Ononis spinosa L. roots (OSR) treatment obesity and metabolic disorders requires а scientific rationale. The current study examined the capacity OSR its secondary metabolites ononin (ONON) maackiain (MACK) in human adipocytes as vitro model obesity. Both ONON MACK diminished lipid accumulation during adipocyte differentiation. Molecular docking analysis exposed potential interactions between target regulatory adipogenic proteins. Furthermore, results from RT-qPCR disclosed significant upregulation AMPK by treatment. In addition, increased SIRT1, PI3K ACC mRNA expression, while notably downregulated CEBPA, AKT, SREBP1, ADIPOQ. protein level PI3K, C/EBPα, PPARγ adiponectin was reduced upon concentration-dependent manner.
