Cytokine licensing with pro-inflammatory molecules, such as tumour necrosis factor-alpha (TNF-α) and interferon-gamma (IFN-γ), has emerged a promising strategy to enhance the therapeutic potential of multipotent mesenchymal stromal cells (MSCs). While demonstrated benefits for immunomodulation, its effects on other key MSC functions, including differentiation paracrine activity, remain incompletely explored. In this study, we evaluated transcriptomic, metabolomic, functional changes induced by short-term TNF-α/IFN-γ priming Wharton's jelly-derived MSCs (WJ-MSCs). WJ-MSCs were expanded exposed TNF-α IFN-γ (10 ng/ml each) 24 h. Transcriptomic analysis was performed using RNA sequencing identify differentially expressed genes related immune modulation lineage commitment. Metabolomic profiling conducted high-resolution mass spectrometry assess in metabolic pathways. Our findings demonstrate that reprograms enhancing their immunomodulatory capacity at expense potential. These results highlight need tailored strategies optimize functionality specific clinical applications.
