Ischemic brain injury results in high disability due to neuroinflammation and oxidative stress, M1/M2 polarization of glial cells plays a key role neuroinflammation. This research explored the protective effect Catalpalactone on middle cerebral artery occlusion (MCAO)-induced its underlying regulation mechanism rats. The ischemic lesions were induced by MCAO, oxygen glucose deprivation/reoxygenation (OGD/R) was used for BV2 microglial cell induction. determined via immunohistochemistry staining assessment. Oxygen consumption rate (OCR) extracellular acidification (ECAR) assays glycolysis phosphorylation test. After that, counting kit-8 (CCK-8) viability test flow cytometry apoptosis analysis performed. Furthermore, co-culture model PC12 purpose exploring effects interaction microglia neurons injury. Finally, Modified Neurological Severity Score (mNSS) neurological function. MCAO induction, infiltration significantly increased area, M1 phenotype enhanced (up-regulated Cd86). In vitro, OGD/R-induced also exhibited increasing with higher activity, but lower through activating JAK-SATA signaling pathway. we that 15 μM optimally induces M2 decreased model, while reducing mNSS scores improving function rat model. We clarified treatment promoting phenotype.

Alzheimer’s disease (AD) is a widespread neurodegenerative disorder and one of the major challenges for public health. Despite extensive research, role microglia in AD remains complex dual. The aim this review to summarize most recent advances research regarding dual concerning both immunomodulation pathological progression by considering mechanisms activation microglia, effects on Aβ clearance, tau pathology, impacts due genetic variations microglial functions. Among these findings are status M1 M2 phenotypes, crucial that variants like TREM2 have modulating response microglia. This describes how modulation signaling pathway might be exploited therapeutically treatment underlines relevance personalized medicine approach.

Background The synergistic epidemic of diabetes mellitus and tuberculosis (DM-TB) has created a dual disease burden, challenging global health systems with complex pathophysiological interactions suboptimal treatment outcomes. To decode the evolving research landscape, this study presents latest comprehensive bibliometric analysis mapping intellectual architecture DM-TB over three decades. Methods We systematically analyzed 791 peer-reviewed articles from Web Science Core Collection (1995-2023) using CiteSpace, VOSviewer, Bibliometrix. Advanced metrics including co-citation networks, keyword burst detection, institutional collaboration patterns were employed to identify paradigm-shifting trends. Results Three distinct growth phases observed: initial stagnation (1995-2007, <10 annual publications), exponential (2008-2019), diversification (2020-2023). United States dominated scientific output (27.3% total while London School Hygiene & Tropical Medicine emerged as central hub for international collaborations (TLS=176). Keyword evolution revealed transformative phases: (1) Pathomechanistic exploration (1995-2016): Focused on hyperglycemia-immunity interplay epidemiological surveillance; (2) Translational innovation (2017-2020): Shifted preclinical models, pharmacokinetic optimization, multidrug resistance; (3) Precision medicine era (2021-2023): Emerging hotspots in latent TB screening (burst strength=6.82), metformin-mediated immunomodulation, AI-driven diagnostic biomarkers. Conclusion Beyond delineating historical trajectories, identifies critical knowledge gaps inflammation-resolution mechanisms insulin resistance pathways, proposing roadmap targeted biomarker discovery policy formulation. constructed framework empowers strategic resource allocation combating syndemic.

Background Traumatic brain injury (TBI) is a critical global health concern characterized by elevated rates of both morbidity and mortality. The pathological physiological changes after TBI are closely related to microglia. Microglia, the primary immune cells in brain, linked mechanisms treatment TBI. With increasing research this area, study employs bibliometric analysis identify current hotspots predict future trends. Objective We decided perform provide comprehensive overview advancements microglia traumatic injury. aim offer researchers insights into trends directions. Method collected all articles reviews published between 2000 2023 from Web Science Core Collection. These records were analyzed using VOSviewer, CiteSpace, R package “bibliometrix”. Results retrieved 665 publications 25 countries, with majority contributed United States China. number on has been steadily each year. Our highlighted Journal Neurotrauma Neuroinflammation as most influential journals field. Alan I. Faden David J. Loane recognized leading contributors. Keyword indicates that neuroinflammation, microglial polarization, neurodegenerative diseases pivotal areas for research. Conclusion In recent years, TBI-related proliferated, studies primarily focusing involvement changes, polarization following Since neuroinflammation neurodegeneration two hallmark features TBI, targeting may become central focus
