Early-onset colorectal cancer (EOCRC) is associated with a poorer prognosis relative to late-onset (LOCRC), and its incidence has witnessed gradual escalation in recent years. This necessitates comprehensive examination of the underlying pathogenesis identification therapeutic targets specific EOCRC patients. The present study aimed delineate distinct molecular landscape by juxtaposing it that LOCRC.

Abstract Aim The existing evidence has shown a positive association between alcohol consumption and an increased risk of colorectal cancer (CRC). However, the is primarily based on studies CRC in all ages, role early onset (EOCRC) remains to be determined. aim this study was investigate increasing incidence EOCRC consumption. Method We systematically searched PubMed, EMBASE, Cochrane Web Science up June 2024 for that evaluated intake with report specific results (e.g. relative risk, OR or hazard ratio corresponding 95% CI). Based varying designs included studies, effect values were extracted categorized into high low groups; random‐effects model adopted estimate pooled sizes analysis. Furthermore, subgroup analyses publication bias assessments conducted. Results Three cohort eight case–control eligible included. meta‐analyses, which yielded heightened (OR = 1.56, CI 1.28–1.89, I 2 89.3%). In analysis, no significant differences found developing across gender, location tumour site. sensitivity analysis indicated conclusion robust. Conclusions This meta‐analysis provides possible EOCRC. More research needed future confirm these findings.

Aim This study aimed to investigate the causes and risk factors of colorectal cancer (CRC) in a Turkish population, focusing on various modifiable non-modifiable factors. Methods A hospital-based case-control design was employed compare individuals with CRC (cases) without (controls). Male female participants were recruited from surgery, internal medicine, out-patient departments. The encompassed socio-demographic data, clinical information, radiological diagnoses, biochemical measurements. Univariable multivariable logistic regressions used determine associated CRC. Results included 704 controls. Significant disparities observed between groups, over 30% cases having lower levels education income compared Lifestyle such as obesity, higher rates smoking (cigarettes hookah) alcohol consumption more prevalent among than Further significant associations identified intestinal inflammation, processed food consumption, symptoms abdominal pain, cramps, diarrhea, constipation, blood stool, bloating, irritable bowel syndrome, nausea/vomiting, anemia, stress, fatigue, weakness, weight loss. Diet analysis revealed that consumed red meat, fast foods along less pulses vegetables. Genetic predispositions exposure chemicals also correlated strongly increased risk. Multivariable regression identified, disease, genetics factor, hookah-nargileh use, history any cancer, family cigarette smoking, milk-yogurt obesity meat determinants for Conclusion is influenced by dietary, lifestyle, genetic Awareness hereditary participation screening are crucial. changes, avoiding hookah, adopting healthy diet, essential prevention.

Abstract Early-onset colorectal cancer (EOCRC) shows a different epidemiological trend compared to later-onset cancer, with its incidence rising in most regions and countries worldwide. However, the reasons behind this remain unclear. The etiology of EOCRC is complex could involve both genetic environmental factors. Apart from Lynch syndrome Familial Adenomatous Polyposis, sporadic exhibits broad spectrum pathogenic germline mutations, polymorphisms, methylation changes, chromosomal instability. Early-life exposures risk factors, including lifestyle dietary have been found be associated risk. Meanwhile, specific chronic diseases, such as inflammatory bowel disease, diabetes, metabolic syndrome, EOCRC. Interactions between factors also explored. Here we present findings narrative review studies on assessment early-life exposures, EOCRC-specific their interactions susceptible loci. We results genome-wide association that used perform Mendelian randomization analyses ascertain potential causal links

PURPOSE In artificial intelligence–based modeling, working with a limited number of patient groups is challenging. This retrospective study aimed to evaluate whether applying synthetic data generation methods the clinical small can enhance performance prediction models. MATERIALS AND METHODS A set collected by Cancer Registry Library Project from Yonsei Center (YCC), Severance Hospital, between January 2008 and October 2020 was reviewed. Patients colorectal cancer younger than 50 years who started their initial treatment at YCC were included. Bayesian network–based synthesizing model used generate set, combined differential privacy (DP) method. RESULTS population 5,005 generated 1,253 patients 93 features. The Hellinger distance correlation difference metric below 0.3 0.5, respectively, indicating no statistical difference. overall survival disease stage did not differ original populations. Training validating showed highest performances 0.850, 0.836, 0.790 for Decision Tree, Random Forest, XGBoost models, respectively. Comparison sets different epsilon parameters improved >0.1%. For extremely sets, models using outperformed those only sets. reidentification risk measures demonstrated that epsilons 0.1 100 fell baseline, preserved state. CONCLUSION approach enhances predictive modeling maintaining integrity, simultaneously reduces risks through application DP techniques.

Background The incidence of early-onset colorectal cancer (EOCRC) rose abruptly in the mid 1990s, is continuing to increase, and has now been noted many countries. By 2030, 25% American patients diagnosed with rectal will be 49 years or younger. large majority EOCRC cases are not found germline susceptibility mutations (eg, Lynch syndrome) inflammatory bowel disease. Thus, environmental lifestyle factors suspected drivers. Obesity, sedentary lifestyle, diabetes mellitus, smoking, alcohol, antibiotics affecting gut microbiome have proposed. However, these factors, which present since 1950s, yet conclusively linked abrupt increase EOCRC. sharp suggests introduction a new risk factor for young people. We hypothesized that driver may an off-target effect pharmaceutical agent (ie, one requiring regulatory approval before its use general population off-label previously approved agent) genetically susceptible subgroup adults.
