ABSTRACT At implantation, with the acquisition of a receptive phenotype in uterine epithelium, an initial tenuous attachment embryonic trophectoderm initiates reorganisation epithelial polarity to enable stable embryo and differentiation invasive trophoblasts. During the implantation window, endometrium becomes poised to transition a pregnant state, process driven by differentiation of stromal cells into decidual (DC). Perturbations in this process, termed decidualization, leads breakdown feto-maternal interface and miscarriage, but underlying mechanisms are poorly understood. Here, we reconstructed pathway at single-cell level vitro demonstrate that first mount an acute stress response before emerging as DC or senescent (snDC). In absence immune cell-mediated clearance snDC, secondary senescence transforms progesterone-resistant abundantly express extracellular matrix remodelling factors. Additional analysis midluteal identified DIO2 SCARA5 marker genes diverging vivo.
