Abstract Colorectal cancer (CRC) ranks among the most prevalent malignancies worldwide, driving a quest for comprehensive characterization methods. We report of ex vivo autofluorescence lifetime fingerprint colorectal tissues obtained from 73 patients that underwent surgical resection. specifically target characteristics collagens, reduced nicotine adenine (phosphate) dinucleotide (NAD(P)H), and flavins employing fiber-based dual excitation (375 nm 445 nm) optical imaging system. Autofluorescence-derived parameters normal tissues, adenomatous lesions, adenocarcinomas were analyzed considering underlying clinicopathological features. Our results indicate differences between are primarily driven by collagen parameters. also changes in associated with NAD(P)H we tentatively attribute to intratumoral heterogeneity, potentially presence distinct metabolic subpopulations. Changes signatures tumors observed lymphatic venous invasion, differentiation grade, microsatellite instability. Finally, characterized impact radiative treatment fingerprints rectal generalized increase mean radiated tumors, which is suggestive altered metabolism structural remodeling. Overall, our preliminary findings multiparametric measurements have potential significantly enhance clinical decision-making CRC, spanning initial diagnosis ongoing management. believe will provide foundational framework future investigations further understand combat CRC exploiting measurements.

ABSTRACT This article gives an overview of the most frequently used fluorescence‐lifetime imaging (FLIM) techniques, their capabilities, and typical applications. Starting from a general introduction to fluorescence phosphorescence lifetime, we will show that lifetime or, more accurately, decay function fluorophore is direct indicator interaction with its molecular environment. FLIM therefore than simple contrast technique in microscopy—it imaging. techniques can be classified into time‐domain frequency‐domain analogue photon counting scanning wide‐field techniques. these technical principles describe features peculiarities different use. An extended section dedicated TCSPC FLIM, addressing unique capabilities make especially interesting biological systems.

Currently, the focus of intraoperative imaging in brain tumor surgery is beginning to shift optical methods such as coherence tomography (OCT), Raman spectroscopy, confocal laser endomicroscopy (CLE), and fluorescence lifetime (FLIM). Optical technologies provide vivo real-time high-resolution images tissues. “Optical biopsy” can be considered an alternative traditional approaches for histopathologic consultation. Intraoperative help achieve precise identification infiltrations within surrounding parenchyma. Therefore, it a complement existing based on wide-field modalities MRI, US, or 5-ALA fluorescence. A promising future direction guidance during stereotactic biopsy lies integration with machine learning techniques, enabling automated differentiation between tissue healthy We present this review increase knowledge form critical opinions field using surgery.
