ABSTRACT Aging induces significant alterations in the immune system, with immunosenescence contributing to age‐related diseases. Peripheral blood mononuclear cells (PBMCs) offer a convenient and comprehensive snapshot of body's status. In this study, we performed an integrated analysis PBMCs using both bulk‐cell single‐cell RNA‐seq data, spanning from children frail elderlies, investigate changes. We observed dynamic changes PBMC transcriptome during healthy aging, including dramatic shifts inflammation, myeloid cells, lymphocyte features early life, followed by relative stability later stages. Conversely, elderly individuals exhibited notable disruptions peripheral increased senescent phenotype monocytes elevated inflammatory cytokine expression, heightened effector activation regulatory T functional impairment cytotoxic lymphocytes. Overall, study provides valuable insights into complex dynamics immunosenescence, elucidating mechanisms driving abnormal inflammation immunosuppression frailty.
