M2-like tumor-associated macrophages (M2-like TAMs) function crucially in the tumor microenvironment (TME) and cancer development. This study developed a prognostic signature based on TAM-related genes for breast (BRCA) applying transcriptome scRNA-seq analysis. TCGA-BRCA, GSE20685, GSE176078 datasets were downloaded from UCSC xena GEO databases. AUCell score of immune-related (IRGs) was calculated using R package. Genes related to TAMs screened by WGCNA. Prognostic further identified univariate Cox LASSO regression analyses form RiskScore model, which validated external dataset. Furthermore, nomogram established integrating clinical characteristics, correlation analysis between TME or chemotherapeutic drugs conducted. Finally, mRNA expression levels key verified quantitative real time polymerase chain reaction (qRT-PCR). As exhibited highest IRGs single-cell transcriptomic atlas BRCA, cells classified into Macrophages C1 C2 subtypes, with subtype showing high M2 macrophage marker genes. ARHGAP26, RILP, KLRB1, CSTA, KLHDC7B, PSMB8, KYNU, RNASE1, LONRF3, TRPM2 as total 903 establish robust model. strong predictive performance constructed combining stage, Age, RiskScore, we found that most immune showed negative RiskScore. Multiple closely associated notably, Ribociclib_1632 had higher half-maximal inhibitory concentration (IC50) value high-risk group. qRT-PCR demonstrated 10 significantly different control BRCA cell lines. Background/Objectives: Nuclear pleomorphism, a crucial determinant of breast cancer grading under the Nottingham Histopathology Grading (NHG) system, remains inadequately quantified in existing literature. Motivated by this gap, our study seeks to investigate and establish correlations among morphological features across various scores nuclear as per NHG system. We aim quantify pleomorphism these validate proposed measurement method against ground-truth data. Methods: Initially, we deconstruct descriptions into three core elements: size, shape, appearance. These elements are subsequently mathematically modeled equations, termed ESize, EShape, EAppearance. equations then integrated unified model Harmonic Mean (HM). The HM equation yields value approaching 1 for nuclei demonstrating characteristics score-3 near 0 those exhibiting score-1 pleomorphism. Results: demonstrates promising performance metrics, including Accuracy, Recall, Specificity, Precision, F1-score, with values 0.97, 0.96, 0.94, 0.95, respectively. Conclusions: In summary, proposes novel feature precise quantification cancer.
