ABSTRACT The transcriptional co-activator Med12 regulates gene expression through the function of its kinase module and by interacting with larger Mediator complex associated RNA Polymerase II (RNAPII). Here, we show a module-independent in antibody class switching recombination (CSR). is essential for IgH 3’ regulatory region (3’RR) or super-enhancer activation functions p300 Jmjd6/Carm1 coactivator complexes. deficiency leads to dramatic decrease H3K27 acetylation enhancer (eRNA) transcription at 3’RR, concomitant impairment AID-induced DNA double strand breaks, long-range S-S synapse formation, 3’RR-Eμ interaction. CRISPR/dCas9-mediated re-establishes epigenomic hallmarks 3’RR super-enhancer, fully restoring depletion defects. Notably, find that derived eRNAs are critical promoting proper S epigenetic regulation, formation recruitment AID locus. We specific X-Linked intellectual disability syndrome mutations defective both eRNA CSR, suggesting B neuronal cells may have cell-specific dysfunctions. conclude IgH3’RR plays central role diversification genomic instability cells.
