Abstract Establishment of implantation in pig is accompanied by a coordinated interaction between the maternal uterine endometrium and conceptus development. We investigated expression profiles endometrial tissue on Days 9, 12 15 pregnancy Day non-pregnancy Yorkshire performed comprehensive analysis long non-coding RNAs (lncRNAs) samples using RNA sequencing. As result, 2805 novel lncRNAs, 2,376 (301 lncRNA 2075 mRNA) differentially expressed genes (DEGs) 2149 transcripts were obtained pairwise comparison. In agreement with previous reports, lncRNAs shared similar characteristics, such as shorter length, lower exon number, at level less conserved than protein coding transcripts. Bioinformatics showed that DEGs involved binding, cellular process, immune system process enriched focal adhesion, Jak-STAT, FoxO MAPK signaling pathway. also found TCONS_01729386 TCONS_01325501 may play vital role embryo pre-implantation. Furthermore, FGF7 , NMB, COL5A3, S100A8 PPP1R3D significantly up-regulated time recognition (Day pregnancy). Our results first identified characterization profile during pre-implantation phases.

Porcine conceptuses secrete pregnancy‐recognition signals (estrogens, including estradiol‐17β) that inhibit luteolysis, thereby prolonging progesterone production by corpora lutea. The supportive mechanism which the conceptus also inhibits luteolysis is shifting endometrial prostaglandin (PG) synthesis to luteoprotective PGE2. Progesterone stimulates of factors are essential for development. Priming uterus and loss receptors from uterine epithelium D1ay 10–12 after estrus key achieving receptivity implantation. Conceptus implantation involves a series events, many resembling inflammatory reaction, greatly influenced cytokines, growth factors, prostaglandins. We herein present novel, dual role PGF2α in lutea depends on acquisition luteolytic sensitivity, based knowledge triggers pathways involved during estrous cycle or/and may have an alternative function maintaining pregnancy. point out new that, together with PGE2, can act as embryonic signal mediators. PGF2α, until recently was considered undesirable promoting pregnancy, now known stimulate conceptus‐maternal interactions angiogenesis endometrium. Polycystic ovary syndrome (PCOS) is one of the most frequent endocrinopathies, affecting 5-10% women reproductive age, and characterized by presence ovarian cysts, oligo, or anovulation, clinical biochemical hyperandrogenism. Metabolic abnormalities such as hyperinsulinemia, insulin resistance, cardiovascular complications, dyslipidemia, obesity are frequently present in PCOS women. Several key pathogenic pathways overlap between these metabolic abnormalities, notably chronic inflammation. The observation that this mechanism was shared led to hypothesis a inflammatory state could contribute pathogenesis PCOS. Moreover, while physiological inflammation an essential feature events ovulation, menstruation, implantation, labor at term, establishment may be pivotal observed dysfunctions Taken together, work aims review available evidence about mediators related mechanisms with PCOS, emphasis on function.

In pigs, conceptus attachment to the uterine surface epithelium starts around Day 14 of pregnancy preceded by a pronounced vascularization at implantation zones, initiating epitheliochorial placentation. To characterize complex transcriptome changes in endometrium course initial attachment, deep sequencing endometrial RNA samples pregnant animals (n = 4) and corresponding cyclic controls was performed using Illumina RNA-Seq. The obtained sequence reads were mapped porcine genome, relative expression values calculated for analysis differential gene expression. Statistical revealed 1933 differentially expressed genes (false discovery rate 1%), 1229 with higher 704 lower mRNA concentration, from animals. Expression selected validated use quantitative real-time RT-PCR. RNA-Seq data compared results microarray study bovine on 18 additional related sets. Bioinformatics concentration strong overrepresentation, particularly immune-related functional terms but also apoptosis cell adhesion. Overrepresented extracellular region, ion transport, adhesion, lipid steroid metabolic process. conclusion, comprehensive differences between highlighted new processes pathways probably involved regulation noninvasive pig.
