DNA-methylation profile scores (MPSs) index biology relevant for lifelong physical and cognitive health, but information on their longitudinal stability in childhood is lacking. Using two waves of data collected from 2014 to 2022 (Mlag between = 2.41 years) N 407 participants (Mage 12.05 years, 51% female, 60% White), test-retest correlations were estimated four salivary MPSs related aging (PhenoAgeAccel, GrimAgeAccel, DunedinPACE), function (Epigenetic-g). varied (test-retest rs 0.38 0.76). did not differ children exposed the COVID-19 pandemic, race-ethnic sex differences apparent. Further research necessary understand which environmental perturbations impact trajectories when are most sensitive those impacts.

Individuals with more education are at lower risk of developing multiple, different age-related diseases than their less-educated peers. A reason for this might be that individuals age slower. There 2 complications in testing hypothesis. First, there exists no definitive measure biological aging. Second, shared genetic factors contribute toward both educational attainment and the development diseases. Here, we tested whether protective effect was associated pace aging after accounting factors.

Epigenetic clocks, quantifying biological age through DNA methylation (DNAmAge), have emerged as potential indicators of brain aging. As the variety DNAmAge algorithms grows, consensus on their efficacy in predicting age-related changes is lacking. This study aimed to explore intricate relationship between diverse and structural cognitive markers Within a cohort 796 elderly patients (mean age, 65.8 ± 7.9 years), we scrutinized 11 algorithms, including Horvath, Hannum, Zhang's PhenoAge, GrimAge, DunedinPACE, principal component (PC)-based PCHorvath, PCHannum, PCPhenoAge, PCGrimAge. We evaluated association with baseline cognition decline, assessed follow-up evaluations at three (T1) six (T2) years postbaseline. Additionally, examined magnetic resonance imaging aging, white matter. clock was best predictor decline memory (β = -0.04) global -0.03), whereas PCGrimAge speed -0.17). The were second-best predictors explaining variability after education (R2 partial 1.66% 2.82%) for 2.13%). PC-trained outperformed respective original version. are strong independent normal population explain additional beyond that accounted by conventional risk factors.

Abstract Translational Geroscience is rooted in the geroscience paradigm which seeks to define genetic, molecular, and cellular mechanisms that make age major risk factor driver of most chronic conditions diseases older people. The final aim convert this fundamental knowledge into effective interventions promote healthy ageing. In chapter, we discuss how interfaces with life course epidemiology through emerging tools assess biological age. It has potential shift focus medicine from current model disease treatment an emphasis on counteracting ageing phenotypes during one’s full expand health expectancy. We claim advances biomarker technologies, science approaches, mechanism-based therapeutic strategies, a day when individuals will be spending their good within sight.

Abstract Background Epigenetic algorithms of aging, health, and cognition, based on DNA-methylation (DNAm) patterns, are prominent tools for measuring biological age have been linked to age-related diseases, cognitive decline, mortality. While most these methylation profile scores (MPSs) developed in blood tissue, there is growing interest using less invasive tissues like saliva. The aim the current study probe cross-tissue intraclass correlation coefficients (ICCs) MPSs applied saliva DNAm from same people. our primary focus that were previously found be robustly correlated with social determinants including second- third-generation clocks physiology we also report ICC values first-generation enable comparison across metrics. We pooled three publicly available datasets had both individuals (total n = 107, aged 5–74 years), corrected cell composition within each computed ICCs. Results after correcting composition, saliva–blood ICCs moderate indices aging cognition. Specifically, PCGrimAge highest (0.76), followed by PCPhenoAge (0.72), a measure performance (Epigenetic- g , 0.69), DunedinPACE (0.68), Acceleration (0.67), (0.66), an MPS hs-CRP (0.58), BMI (0.54). These appear lower than previous reports within-tissue (saliva range 0.67 0.85, 0.73 0.93). Cross-tissue acceleration measures poor, ranging 0.19 0.25. Conclusions Our findings suggest applying related phenotypes results similarity precise correspondence differs measure. degree several may suffice some research settings, it not suitable clinical or commercial applications. Abstract Objectives Loneliness may influence aging biomarkers related to cognitive functioning, for example, through accelerated DNA methylation (DNAm) aging. Methods In the present study, we tested whether six common DNAm age acceleration measures mediated effects of baseline loneliness and five different longitudinal trajectories on general ability, immediate memory recall, delayed processing speed in 1,814 older adults Health Retirement Study. Results We found that individuals who belong highest had poorer ability scores. Only index physiological comorbidities, unhealthy lifestyle factors (e.g., smoking), mortality risk-mediated functioning but not speed. These same moderate-but-declining trajectory functioning. Additionally, scores were by GrimAge Accel lowest two groups. Total outcomes mainly accounted demographic, social, psychological, covariates, most notably self-rated health, depressive symptomatology, objective social isolation, body mass index. Discussion Current findings suggest aging, particularly Accel, have promise explaining prospective association between outcomes.
