Abstract Regulated cell death (RCD) is a regulable that involves well-organized signaling cascades and molecular mechanisms. RCD implicated in fundamental processes such as organ production tissue remodeling, removing superfluous structures or cells, regulating numbers. Previous studies have not been able to reveal the complete mechanisms, novel methods of are constantly being proposed. Two metal ions, iron (Fe) copper (Cu) essential factors leading RCDs only induce ferroptosis cuproptosis, respectively but also lead impairment eventually diverse death. This review summarizes direct indirect mechanisms by which Fe Cu impede growth various forms mediated these two metals. Moreover, we aimed delineate interrelationships between with distinct pathways shedding light on complex intricate govern cellular survival Finally, prospects outlined this suggest approach for investigating death, may involve integrating current therapeutic strategies offer promising solution overcome drug resistance certain diseases.

Type 2 diabetes (T2D) and Clear-cell renal cell carcinoma (ccRCC) are both complicated diseases which incidence rates gradually increasing. Population based studies show that severity of ccRCC might be associated with T2D. However, so far, no researcher yet investigated about the molecular mechanisms their association. This study explored T2D causing shared key genes (sKGs) from multiple transcriptomics profiles to investigate common pathogenetic processes drug molecules. We identified 259 differentially expressed (sDEGs) can separate patients control samples. Local correlation analysis on expressions sDEGs indicated significant association between ccRCC. Then ten (CDC42, SCARB1, GOT2, CXCL8, FN1, IL1B, JUN, TLR2, TLR4, VIM) were selected as sKGs through protein-protein interaction (PPI) network analysis. These found significantly different CpG sites DNA methylation cause The sKGs-set enrichment Gene Ontology (GO) terms KEGG pathways revealed some crucial functions, biological process, cellular components development regulatory six post-transcriptional regulators (hsa-mir-93-5p, hsa-mir-203a-3p, hsa-mir-204-5p, hsa-mir-335-5p, hsa-mir-26b-5p, hsa-mir-1-3p) five transcriptional (YY1, FOXL1, FOXC1, NR2F1 GATA2) sKGs. Cuproptosis and ferroptosis represent copper- iron-dependent forms of cell death, respectively, both are known to play pivotal roles in head neck squamous carcinoma (HNSCC). However, few studies have explored the prognostic signatures related cuproptosis HNSCC. Our objective was construct a model based on genes associated with ferroptosis. We randomly assigned 502 HSNCC samples from The Cancer Genome Atlas (TCGA) into training testing sets. Pearson correlation analysis utilized identify cuproptosis-associated set. Cox proportional hazards (COX) regression least absolute shrinkage operator (LASSO) were employed model. performance internally validated using single-factor COX regression, multifactor Kaplan–Meier analysis, principal component (PCA), receiver operating curve (ROC) analysis. Additionally, we obtained 97 Gene Expression Omnibus (GEO) database for external validation. constructed model, 12 genes, proved be an independent predictor HNSCC prognosis. Among these increased expression aurora kinase A (AURKA) has been implicated various cancers. To further investigate, small interfering RNAs (siRNAs) knock down AURKA conducted functional experiments. results demonstrated that knockdown significantly inhibited proliferation migration cells (Cal27 CNE2). Regulated cell death (such as apoptosis, necroptosis, pyroptosis, autophagy, cuproptosis, ferroptosis, disulfidptosis) involves complex signaling pathways and molecular effectors, has been proven to be an important regulatory mechanism for regulating neuronal aging death. However, excessive activation of regulated may lead the progression aging-related diseases. This review summarizes recent advances in understanding seven forms age-related Notably, newly identified ferroptosis cuproptosis have implicated risk cognitive impairment neurodegenerative These exacerbate disease by promoting inflammation, oxidative stress, pathological protein aggregation. The also provides overview key crosstalk mechanisms among these forms, with a focus on disulfidptosis. For instance, FDX1 directly induces copper ion valency dihydrolipoamide S-acetyltransferase aggregation, while mediates glutathione peroxidase 4 degradation, enhancing sensitivity. Additionally, inhibiting Xc- transport system prevent can increase disulfide formation shift NADP + /NADPH ratio, transitioning insights help uncover potential connections novel differentiate them from traditional mechanisms.
