Abstract Background: PANoptosis, a recently identified pathway of cell death, entails interplay and coordination among pyroptosis, apoptosis, necroptosis mechanisms. Yet, the prognostic implications PANoptosis-related genes (PRGs) in clear renal carcinoma (ccRCC) remain largely unexplored. Methods: In this study, nineteen were from prior research, clinical data patients with retrieved TCGA. training cohort, we conducted univariate Cox, Lasso, multivariate Cox regression analyses to identify subsequently develop PRGs signature. The signature was validated using both testing cohort entire cohort. Furthermore, nomogram developed predict prognosis at various clinicopathological stages risk scores. Additionally, network comprising 6 prognosis-related differentially expressed (PRDEGs) 5 predicted functional partners constructed. Results: We utilized analysis assess significance 19 (PRGs). Subsequently, employed least absolute shrinkage selection operator (Lasso) further evaluate value these PRGs. established model consisting six PRDEGs: TAB2, TAB3, TNFAIP3, CASP7, AIM2, TRADD. Kaplan-Meier (KM) time-related receiver operating characteristic (ROC) curve performed overall survival compare high- low-risk groups. Incorporating characteristics, as novel independent factor for through analyses. demonstrated reliable predictive ability. Conclusion: PRDEGs showed notable ccRCC. These hold promise potential therapeutic targets settings.
