Global economic and social burden was caused by the SARS-CoV-2 spread worldwide. Despite end of pandemic, there is a concern about virulent evolving variants virus which can bypass humoral immune response induced vaccination or infection. Crucial to viral entrance, amino acid residues in RBM region, interacts with cellular receptor ACE2, elicit neutralizing antibody response. Herein we determine immunogenicity one-dose heterologous dose vaccinated serum against wild-type mutated region. low RBM, omicron possess four mutations (S477N, T478K, E484A, F486V) that induce even less titers. The most predominant responses were IgA IgG. While antibodies (nAbs) predominantly target RBD, our investigation revealed diminished seroreactivity within RBD's crucial receptor-binding motif (RBM), potentially impacting production protective nAbs. S1WT S2WT peptides binding nAbs evaluated through microscale thermophoresis, higher affinity (35 nM) obtained for sequence (GSTPCNGVEGFNCYF), containing FNCY patch. Our data indicates not an immunodominant region individuals. Understanding intricate dynamics its interplay evolution host genetics essential formulation effective strategies, only but also future emerging coronaviruses.

The newly introduced COVID-19 vaccines have reduced disease severity and hospitalizations. However, they do not significantly prevent infection or transmission. In the same context, measuring IgM IgG antibody levels is important, but it does provide information about status of mucosal immune response. This article describes a comprehensive mapping IgA epitopes S protein, its cross-reactivity, development an ELISA-peptide assay.IgA epitope was conducted using SPOT synthesis sera from RT-qPCR COVID-19-positive patients. Specific cross-reacting were identified, evolutionary analysis early Wuhan strain to Omicron variant performed bioinformatics tools microarray peptides. selected chemically synthesized evaluated ELISA-IgA.A total 40 identified with 23 in S1 17 S2 subunit. Among these, at least showed cross-reactivity DENV other organisms 24 associated coronaviruses. Three MAP4 polypeptides validated by ELISA, demonstrating sensitivity 90-99.96% specificity 100%. six IgA-RBD epitopes, only SC/18 variants (BA.2 BA.2.12.1) presented single epitope. This research unveiled epitome protein many that exhibit retains conserved except for one (#SCov/18). suggests limitations whole S1/S2/RBD segment serological diagnostic tests COVID-19. expression these specific as biomarkers could facilitate monitoring immunity COVID-19, potentially leading more accurate diagnoses alternative vaccines.
