Alzheimer’s disease and other neurodegenerative disorders are characterized by the accumulation of misfolded proteins, such as amyloid-beta, tau, α-synuclein, which disrupt neuronal function contribute to cognitive decline. Heparan sulfate proteoglycans, particularly syndecans, play a pivotal role in seeding, aggregation, spreading toxic protein aggregates through endocytic pathways. Among these, syndecan-3 is critical regulating internalization facilitating their propagation prion-like manner. This review examines mechanisms especially SDC3, seeding pathological diseases. Understanding these pathways provides valuable insights into potential syndecans biomarkers therapeutic targets for early intervention related disorders.
