Danshen has been widely used for the treatment of central nervous system diseases. We investigated effect dihydroisotanshinone I (DT), a compound extracted from Danshen, as well corresponding mechanisms in an vitro-based 6-OHDA-induced Parkinson's disease (PD) model. SH-SY5Y human neuroblastoma cell lines were pretreated with 6-hydroxydopamine (6-OHDA) and challenged DT. Subsequently, viability levels reactive oxygen species (ROS) caspase-3 analyzed. The DT on 6-OHDA-treated cells expression core circadian clock genes measured using real-time quantitative polymerase chain reaction. Our results indicated that attenuated death suppressed ROS caspase-3. Moreover, reversed both RNA protein BMAL1 SIRT1 cells. Additionally, inhibitor reduced viability. activators activated BMAL1, then damaged by 6-OHDA. silencing was enhanced resulted downregulation reduction In conclusion, our investigation suggested reduces apoptosis, including antioxidative due to ROS, regulates enhancing suppressing BMAL1. may possess novel therapeutic potential PD future, but further vivo studies are still needed.

Gel electrophoresis and size exclusion chromatography (SEC) are vital techniques in biochemical research, employing gel matrix structures made of polysaccharides or synthetic polymers like polyacrylamide for the analysis separation macromolecules. Polysaccharides, such as agarose, offer safer alternatives to acrylamide. Polysaccharide gels, notably facilitate purification proteins nucleic acids through a molecular sieving mechanism. is mainly divided into denaturing native methods. Denaturing with sodium dodecyl sulfate (SDS) simplifies protein migration but disrupts interactions. Conversely, electrophoresis, without SDS, allows migrate based on running pH isoelectric point proteins, while consistently toward anode. The variable charges presents complexes. This review focuses use polysaccharides, particularly highlighting their applications separating It also discusses limitations agarose gels when used electrophoresis. Such information should help designing experiments using polysaccharides.

Sarcoma is a rare and complex disease comprising over 80 malignant subtypes that frequently characterized by poor prognosis. Challenges in clinical management include uncertainties diagnosis classification, limited prognostic predictive biomarkers, incompletely understood heterogeneity among within subtypes, lack of effective treatment options, progress identifying new drug targets novel therapeutics. Proteomics refers to the study entire complement proteins expressed specific cells or tissues. Advances proteomics have included development quantitative mass spectrometry (MS)-based technologies which enable analysis large numbers with relatively high throughput, enabling be studied on scale has not previously been possible. Cellular function determined levels various their interactions, so offers possibility insights into cancer biology. therefore potential address some key current challenges described above, but it still its infancy. This review covers proteomic sarcoma studies findings pertain utility. Proteomic methodologies applied human research are briefly described, including recent advances MS-based technology. We highlight illustrate how may aid improve classification distinguishing histologies identify distinct profiles histological understanding heterogeneity. also where prognostic, therapeutic biomarkers. These traverse range chordoma, Ewing sarcoma, gastrointestinal stromal tumors, leiomyosarcoma, liposarcoma, peripheral nerve sheath myxofibrosarcoma, rhabdomyosarcoma, synovial osteosarcoma, undifferentiated pleomorphic sarcoma.
