Abstract Melanoma, renowned for its aggressive behavior and resistance to conventional treatments, stands as a formidable challenge in the oncology landscape. The dynamic complex interplay between cancer cells tumor microenvironment has gained significant attention, revealing Melanoma-Associated Fibroblasts (MAFs) central players disease progression. heterogeneity of MAFs endows them with dual role melanoma. This exhaustive review seeks not only shed light on multifaceted roles orchestrating tumor-promoting inflammation but also explore their involvement antitumor immunity. By unraveling novel mechanisms underlying MAF functions, this aims provide comprehensive understanding impact melanoma development. Additionally, it delves into potential leveraging innovative immunotherapeutic strategies, offering new avenues enhancing treatment outcomes challenging realm therapeutics.

Lung adenocarcinoma (LUAD) poses significant challenges due to its complex biological characteristics and high recurrence rate. The rate of LUAD is closely associated with cellular dormancy, which enhances resistance chemotherapy evasion immune cell destruction. Using single-cell RNA sequencing (scRNA-seq) data from patients, we categorized the cells into two subclusters: dormant active cells. Utilizing high-density Weighted Gene Co-expression Network Analysis (hdWGCNA) pseudo-time trajectory, aberrant expression genes involved in protein O-glycosylation was detected cells, suggesting a crucial role for maintaining state. Intercellular communication analysis highlighted interaction between fibroblasts where Insulin-like Growth Factor (IGF) signaling pathway regulated by crucial. By employing Set Variation (GSVA) machine learning, risk score model developed using hub genes, showed accuracy determining prognosis. also demonstrated robust performance on training dataset excellent predictive capability, providing reliable basis predicting patient clinical outcomes. group higher exhibited propensity adverse outcomes tumor microenvironment (TME) mutational burden (TMB). Additionally, 50% inhibitory concentration (IC50) values variations among different groups. In vitro experiments that EFNB2, PTTG1IP, TNFRSF11A were upregulated contributed greatly diagnosis LUAD. conclusion, this study dormancy state tumors prognosis patients.
