The tumor microenvironment (TME) is a well-recognized system that plays an essential role in initiation, development, and progression. Intense intercellular communication between cells other (especially macrophages) occurs the TME mediated by cell-to-cell contact and/or soluble messengers. Emerging evidence indicates noncoding RNAs (ncRNAs) are critical regulators of relationship within TME. In this review, we provide update on regulation ncRNAs (primarily micro [miRNAs], long [lncRNAs], circular [circRNAs]) crosstalk macrophages hepatocellular carcinoma (HCC). These derived from or act as oncogenes suppressors, contributing to progression not only regulating physiological pathological processes but also controlling macrophage infiltration, activation, polarization, function. Herein, explore options available for clinical therapeutic strategies targeting crosstalk-related treat HCC. Abstract Targeted treatment, which can specifically kill tumour cells without affecting normal cells, is a new approach for therapy. However, tend to acquire resistance targeted drugs during treatment. Circular RNAs (circRNAs) are single-stranded RNA molecules with unique structures and important functions. With the development of sequencing technology, circRNAs have been found be widespread in tumour-resistant play regulatory roles. In this review, we present latest advances circRNA research summarize various mechanisms underlying their regulation. Moreover, review role chemotherapeutic tumours explore clinical value regulation treating resistance.

Abstract Background Recent studies have revealed that circular RNAs (circRNAs) play significant roles in the occurrence and development of many kinds cancers including breast cancer (BC). However, potential functions most circRNAs molecular mechanisms underlying progression BC remain elusive. Method Here, Circular RNA microarray was executed 4 pairs tissues para-cancer tissues. The expression prognostic significance circACTN4 cells were determined by qRT-PCR situ hybridization. Gain-and loss-of-function experiments implemented to observe impacts on growth, invasion, metastasis vitro vivo. Mechanistically, chromatin immunoprecipitation, luciferase reporter, pulldown, mass spectrum, fluorescence hybridization co-immunoprecipitation assays executed. Results CircACTN4 significantly upregulated cells, its correlated with clinical stage poor prognosis patients BC. Ectopic strikingly facilitated Whereas knockdown opposite roles. mainly distributed nucleus. Further mechanistic research proved could competitively bind far upstream element binding protein 1 (FUBP1) prevent combination between FUBP1 FIR, thereby activating MYC transcription facilitating tumor cancer. Furthermore, we found factor 2 (USF2) might promote biogenesis circACTN4. Conclusion Our findings uncover a pivotal mechanism mediated USF2 interact via enhancing MYC. be novel target for diagnosis treatment

Abstract The role of circular RNAs in oncogenesis has begun to be widely studied recent years, due the significant impact that these molecules have disease pathogenesis, as well their potential for future innovative therapies. Moreover, characteristically shape, are very resistant RNA degradation whose levels easily assessed body fluids. Accordingly, they represent an opportunity discovery new diagnostic and prognostic markers a wide range diseases. Among RNAs, circPVT1 is rather peculiar one originates from circularization exon 2 PVT1 gene encodes pro-tumorigenic long non-coding named lncPVT1. There few examples derive locus producing another RNA. Despite apparent transcriptional independence, which occurs using two different promoters, possible synergistic effect tumorigenesis cannot excluded considering both been reported correlate with oncogenic phenotype. This complex mechanism regulation appears also controlled by c-MYC. Indeed, located only 53 Kb downstream c-MYC gene, well-known oncogene regulates expression about 15% all genes. Here, we review origin biogenesis highlighting most important mechanisms through it plays fundamental oncogenesis, such sponge activity on microRNAs, its paradigmatic interactome link lncPVT1 expression.

Specifically, we detail how inhibits aerobic glycolysis downregulating key glycolytic enzymes metabolic pathways, thereby reducing energy supply essential for cell proliferation. We examine suppresses angiogenesis targeting vascular endothelial growth factor (VEGF) cyclooxygenase-2 (COX-2) effectively starving tumors nutrients oxygen required metastasis. Furthermore, modulates immune microenvironment enhancing activity effector cells such as CD4 + CD8 T natural killer (NK) while immunosuppressive like regulatory (Tregs) myeloid-derived suppressor (MDSCs).
