To explore the differences in protein quality among classic medicinal entomopathogenic fungi and to evaluate their metabolic adaptability, we analyzed amino acid composition proteomic characteristics of Cordyceps sinensis (CS), militaris (CM), cicadae (CC). Quantitative analysis showed CM contained highest crude lysine, methionine, threonine, valine. CS adapted high-altitude hypoxia exhibited lower but elevated leucine, isoleucine, histidine contents, which may contribute membrane stabilization oxidative stress resistance. CC displayed higher non-essential acids such as arginine, proline, tyrosine, reflecting active nitrogen metabolism. Four-dimensional data-independent acquisition (4D-DIA) proteomics identified 495 differentially expressed proteins (DEPs). Compared with CS, upregulated glutamate oxaloacetate transaminases 2 (GOT2), dehydrogenase (GDH), argininosuccinate synthase 1 (ASS1) coordinately regulate flux through alanine-aspartate-glutamate network urea cycle, supporting intermediate replenishment for energy The upregulation branched-chain keto E1 subunit alpha (BCKDHA) acyl-CoA short/branched chain (ACADSB) facilitated integration catabolism TCA explaining species-specific content.
