Abstract Background Alzheimer's disease (AD) is a common type of dementia characterized by amyloid-β (Aβ) accumulation, lysosomal dysfunction, and tau hyperphosphorylation, leading to neurite dystrophy memory loss. This study aimed investigate whether Rhei Undulati Rhizoma (RUR), which has been reported have anti-neuroinflammatory effect, attenuates Aβ-induced impairment, neuritic dystrophy, reveal its mode action. Methods Five-month-old 5xFAD mice received RUR (50 mg/kg) orally for 2 months. The Y-maze test was used assess working memory. After behavioral testing, brain tissue analyzed using thioflavin S staining, western blotting, immunofluorescence staining the action RUR. To confirm directly reduces Aβ aggregation, T assay dot blot were performed after incubating with Results administration attenuated impairment in mice. Furthermore, decreased accumulation observed hippocampus RUR-treated group compare vehicle-treated group. Moreover, reduced dystrophic neurites (DNs) that accumulate impaired endolysosomal organelles around Aβ.
