Abstract Background Hepatocellular carcinoma (HCC) is a leading cause of cancer-related deaths globally, influenced by aberrant circRNA expression. Investigating circRNA-miRNA-mRNA interactions can unveil underlying mechanisms HCC and identify potential therapeutic targets. Methods In this study, we conducted differential analyses mRNAs, miRNAs, circRNAs, established their relationships using various databases such as miRanda, miRDB, miTarBase. Additionally, functional enrichment immune infiltration were performed to evaluate the roles key genes. We also qPCR assays western blotting (WB) examine expression levels circRNA, CCL25, MAP2K1 in both cells clinical samples. Furthermore, utilized overexpression knockdown techniques for circ_0000069 wound healing, transwell invasion assays, tumorigenesis experiment assess migratory invasive abilities cells. Results Our findings revealed significant 612 upregulated genes 1173 downregulated samples compared normal liver tissue. 429 circRNAs 453 identified. Significantly, exhibited upregulation tissues cell lines. The notably increased migration capacity Huh7 cells, whereas downregulation reduced capability HepG2 effect was counteracted CCL25 silencing or overexpression, separately.
