Abstract In the intricate landscape of cellular communication, small extracellular vesicles (sEV) originating from endosomes play crucial roles as mediators and have garnered significant attention in theranostics. Increasing antimicrobial resistance (AMR) challenges conventional antibiotics, prompting the search for alternatives. Extracellular vesicles (EVs) from pasteurised cattle milk offer promise, due to their unique properties. This study investigates efficacy against five pathogenic bacteria, including Staphylococcus aureus ATCC 25923, aiming combat AMR and develop new therapies. EVs were characterised tested using various methods. Co-culture experiments with S. showed significant growth inhibition, colony-forming units decreasing 2.4 × 105 CFU/mL (single dose) 7.4 104 (triple doses) after 12 h. Milk extended lag time (6 9 h) increased generation (2.8 4.8 dose-dependently, compared controls. In conclusion, exhibit dose-dependent inhibition aureus, prolonging times. Despite limitations, this suggests potential in addressing AMR.

Abstract Extracellular vehicles ( EVs ) are an emerging class of drug carriers and primarily reported to be internalized into recipient cells via a combination endocytic routes such as clathrin-mediated, caveolae-mediated macropinocytosis pathways. In this work, 1 we investigated potential effects homotypic vs . heterotypic interactions by studying the cellular uptake homologous (EV donor same type) heterologous different types) 2 determined route EV internalization low pinocytic/hard-to-deliver cell models brain endothelial BECs ). We used macrophages low-pinocytic phagocytic models, respectively, study effect on in cells. Homotypic led greater extent compared interactions. However, did not see complete reduction when pathways were blocked using pharmacological inhibitors. Our results suggest that use membrane fusion enter instead relying endocytosis. Lipophilic PKH67 dye-labeled but intravesicular esterase-activated calcein ester-labeled severely reduced particle while both types EV-labeled particles comparable extents. also highlight importance carefully choosing labeling dye chemistry uptake, especially case pinocytic BECs.
