Being exposed to childhood or gender-based violence is associated with subsequent adverse events in individual lives. Not only can it cause psychological distress but survivors suffer from a range of long-term health outcomes, including higher morbidity, mortality, and risk chronic diseases. Abstract The constant decline in fertility and older reproductive age is the major cause of low clinical pregnancy rates industrialised countries. Epigenetic mechanisms impact on proper embryonic development women undergoing vitro fertilisation (IVF) protocols. Here, we describe main epigenetic modifications that may influence female reproduction could affect IVF success. Graphical

Biological age, reflecting the cumulative damage in body over a lifespan, is dynamic measure more indicative of individual health than chronological age. Accelerated aging, when biological age surpasses implicated poorer clinical outcomes, especially for breast cancer (BC) survivors undergoing treatments. This preliminary study investigates impact 16-week online supervised physical activity (PA) intervention on post-surgery female BC patients. Telomere length was measured using qPCR, and ELOVL2-based epigenetic clock assessed via DNA methylation pyrosequencing

ABSTRACT Premature ovarian insufficiency (POI) has recently been reported to be linked with epigenetic changes. Previous studies have focused on the regulation of individual genes associated function through single‐gene variations; however, there is a deficiency in comprehensive comprehension profile for POI. Therefore, we conducted multi‐omics study integrating methylation, hydroxymethylation and transcriptome sequencing analyses cumulus cells from women POI their matched controls. Our data revealed significant global increases methylation levels patients. We observed predominance hypermethylated hyperhydroxymethylated regions across genome, gene bodies negatively correlating expression, especially promoter regions. Subsequent experimental validation was performed confirm involvement candidate (EGR1, EGR2 DLX5) steroid hormone synthesis. Interestingly, our findings indicate that these modifications are implicated POI, age clock. This underscores potential identifying novel biomarkers therapeutic targets by unravelling complex interplay between DNA epigenetics function.

Quantifying Tissue Response after Intraovarian Platelet-Rich Plasma: Experimental Approach to Assess NF-κB and TNF-α as Mediators to Reduce Local Entropy DOI Open Access
E. Scott Sills

Published: Aug. 14, 2023

Diminished ovarian reserve can be regarded as a sentinel indicator to foreshadow severe follicular loss and, ultimately, systemic aging. The negative slope of human ovulatory fidelity begins with robust endowment which gradually declines over time. In contrast, the youthful phenotype requires coordinated work endothelial, granulosa, immune, perivascular, stromal and possibly germline stem cells. Such diverse tissue matrix can, in general, influenced by platelet (PLT)-derived factors but this has not yet been specifically confirmed ovary after platelet-rich plasma (PRP). How could comparable response validated? Here prospective, experimental study is proposed whereby eligible patients already undergoing scheduled laparoscopy provide via biopsy submitted for co-culture autologous Ca+2 activated PRP. Recognizing interlocking, central roles nuclear factor κB (NF-κB) tumor necrosis factor-α (TNF-α), incubated samples would assessed these vitro before vs. PRP exposure, addition stereomicroscopy. A mathematical model available track NF-κB oscillations estimate gene expression, cell development, growth, apoptosis, key immune inflammatory actions. Since TNF-α are discharged PLT releasate (or react its cargo proteins) audit permits extraction markers observed post-stimulus, thus linking discrete signals transcriptional output, cellular fitness, cytoarchitecture. From this, hypothesis emerge where intraovarian found make no direct impact on follicles, although modified field function curtailed local entropy incidentally favor optimized oocyte competence secondary effect.
