Abstract Long non-coding RNAs (lncRNAs) play an important role in ovarian aging by affecting the biological functions of granulosa cells (GCs) through multiple mechanisms. The lncRNA NEAT1 is significantly downregulated ovaries; however, exact regulatory mechanism has not been elucidated. In current study, we aimed to investigate effects GC during and explore its therapeutic potential. We observed that expression GCs older patients with diminished reserve (DOR), which closely associated function assisted reproductive cycle outcomes. Functional assays revealed promotes KGN cell proliferation increasing proportion S-phase inhibiting apoptosis. Bioinformatics analysis a dual-luciferase reporter assay confirmed acts as miR-204-5p sponge identified ESR1 target gene, both were differentially expressed DOR. Mechanistic experiments demonstrated competitive endogenous RNA adsorbs molecular sponging, turn upregulates key enzymes (steroidogenic acute protein cytochrome P450 family 19 subfamily A member 1) involved steroid hormone synthesis. This induces estradiol biosynthesis activates downstream mitogen-activated kinase (MAPK) signaling pathway promote phosphorylation extracellular signaling-related cyclic adenosine monophosphate response element binding protein, affects results promotion inhibition apoptosis cells. Our suggest MAPK miR-204-5p/ESR1 axis; regulates proliferation, apoptosis, cycle; biosynthesis. Therefore, represents potential delay aging.

PCAT19, a long non-coding RNA, has attracted considerable attention due to its diverse roles in various malignancies. This work compiles current research on PCAT19’s involvement cancer pathogenesis and progression. Abnormal expression of PCAT19 been observed cancers, correlation with clinical features prognosis positions it as promising prognostic biomarker. Additionally, ability effectively differentiate between tumor normal tissues suggests significant diagnostic value. exhibits dual nature, functioning either an oncogene or suppressor, depending the type. It is implicated range tumor-related activities, including cell proliferation, apoptosis, invasion, migration, metabolism, well growth metastasis. acts competing endogenous RNA (ceRNA) interacts proteins regulate critical cancer-related pathways, such MELK signaling, p53 cycle pathways. Furthermore, emerging evidence that plays role modulation neuropathic pain, adding complexity functional repertoire. By exploring molecular mechanisms pathways associated we aim provide comprehensive understanding multifaceted human health disease, highlighting potential therapeutic target for pain management.
