Abstract Neurodegenerative disorders, including Alzheimer’s disease (AD), Parkinson’s (PD), multiple sclerosis (MS), and amyotrophic lateral (ALS), are characterized by the progressive gradual degeneration of neurons. The prevalence rates these disorders rise significantly with age. As life spans continue to increase in many countries, number cases is expected grow foreseeable future. Early precise diagnosis, along appropriate surveillance, continues pose a challenge. high heterogeneity neurodegenerative diseases calls for more accurate definitive biomarkers improve clinical therapy. Cell-free DNA (cfDNA), fragmented released into bodily fluids via apoptosis, necrosis, or active secretion, has emerged as promising non-invasive diagnostic tool various diseases. cfDNA can serve an indicator ongoing cellular damage mortality, neuronal loss, may provide valuable insights processes, progression, therapeutic responses. This review will first cover key aspects then examine recent advances its potential use biomarker disorders.

Cell-free DNA (cfDNA) analysis is a pivotal tool in non-invasive diagnostics, including cancer monitoring and prenatal testing. However, the preanalytical phase, particularly choice of anticoagulant, significantly impacts cfDNA integrity yield. This study aims to compare yield, stability, DNase activity plasma-citrate plasma-heparin, using plasma-EDTA serum as established controls, explore more deeply impact blood DNAse on these specimens. Blood samples from 15 healthy volunteers were collected four types tubes (citrate, heparin, EDTA, serum). was extracted quantified qPCR, endogenous assessed through hydrolysis probe assays. Samples incubated at 37 °C for 24 h evaluate degradation rates. Heparin-plasma exhibited highest activity, with baseline levels intermediate—higher than EDTA but lower serum—leading substantial (85.3%). Combined its known PCR inhibition, this renders heparin-plasma unsuitable analysis. Abstract Circulating cell‐free DNA (cfDNA) is a promising biomarker for physiological stress, including exercise‐induced responses. However, the lack of standardization in blood collection tubes (BCTs) quantification cfDNA hampers inter‐study comparisons. In this study, we assessed impact different BCTs on dynamics.
