Cell-free DNA (cfDNA) analysis is a pivotal tool in non-invasive diagnostics, including cancer monitoring and prenatal testing. However, the preanalytical phase, particularly choice of anticoagulant, significantly impacts cfDNA integrity yield. This study aims to compare yield, stability, DNase activity plasma-citrate plasma-heparin, using plasma-EDTA serum as established controls, explore more deeply impact blood DNAse on these specimens. Blood samples from 15 healthy volunteers were collected four types tubes (citrate, heparin, EDTA, serum). was extracted quantified qPCR, endogenous assessed through hydrolysis probe assays. Samples incubated at 37 °C for 24 h evaluate degradation rates. Heparin-plasma exhibited highest activity, with baseline levels intermediate—higher than EDTA but lower serum—leading substantial (85.3%). Abstract Circulating cell‐free DNA (cfDNA) is a promising biomarker for physiological stress, including exercise‐induced responses. However, the lack of standardization in blood collection tubes (BCTs) quantification cfDNA hampers inter‐study comparisons. In this study, we assessed impact different BCTs on dynamics.
