Background: Female reproductive aging remains irreversible. More evidence is needed on how polyunsaturated fatty acids (PUFAs) affect the female lifespan. Objectives: To identify and validate specific PUFAs that can influence timing of menarche menopause in women. Methods: We utilized a two-sample Mendelian randomization (MR) framework to evaluate causal relationships between various longevity, defined by age at (AAM) natural (ANM). Our analyses leveraged summary statistics from four genome-wide association studies (GWASs) plasma concentrations 10 PUFAs, including 8866 121,633 European individuals 1361 East Asian individuals. Large-scale GWASs for traits provided genetic data AAM ANM over 202,323 females 43,861 females. Causal effects were estimated beta coefficients, representing, each increase standard deviation (SD) PUFA concentration, yearly or ANM. Replications, meta-analyses, cross-ancestry assessed inference. Conclusions: Higher DHA was identified be associated with delayed without affecting menarche, offering potential intervention target extending longevity.

There is growing global concern about the shift to earlier onset of puberty among girls; however, prevalences precocious and normal variant Thai girls are uncertain. We conducted a study at King Chulalongkorn Memorial Hospital in Bangkok from 2018 2022 (amid coronavirus disease 2019 pandemic) investigate prevalence, characteristics, treatment early large cohort girls. The medical records 583 who had been evaluated for were reviewed. Demographic clinical approaches analyzed. analyses found significant annual increase evaluations signs (P-value trend 0.04). majority (71.2%) cases categorized as gonadotropin-releasing-hormone (GnRH)-dependent or central (CPP), with 16 showing abnormal brain magnetic resonance imaging findings. Among puberty, was most common category (10.5%), followed by premature thelarche (9.5%), slow-progressive (6.6%), adrenarche (2.2%). Median age presentation, basal luteinizing hormone (LH) follicle-stimulating (FSH), bone differed significantly groups. Importantly, 97% CPP received GnRH agonists. Girls exhibited higher body mass index z-scores LH FSH compared those puberty. Our highlighted impact obesity on pubertal timing. findings underscore need preventive weight management country-specific guideline updates enhance healthcare strategies young

The increasing global incidence of precocious puberty, linked to environmental, metabolic, and genetic factors, necessitates innovative therapies beyond gonadotropin-releasing hormone (GnRH) analogs. Accumulating evidence implicates gut microbiota dysbiosis as a pivotal regulator pubertal timing via interactions with metabolism (e.g., estrogen reactivation β -glucuronidase), neuroendocrine pathways (nitric oxide signaling), immune-inflammatory responses. This review delineates taxonomic alterations in central puberty (CPP) obesity-related subtypes, including Streptococcus enrichment Alistipes depletion, alongside functional shifts microbial metabolite production. Mechanistic insights highlight microbiota-driven modulation the hypothalamic–pituitary-gonadal (HPG) axis, leptin/insulin dynamics, epigenetic regulation. Emerging interventions-probiotics, fecal transplantation (FMT), dietary modifications-demonstrate efficacy preclinical models early clinical studies for delaying onset restoring hormonal balance. Translational efforts validate these strategies are critical addressing psychosocial challenges posed by positioning novel therapeutic frontier pediatric endocrinology.

Abstract Objective Recent studies suggest that boys enter puberty at a younger age, and the incidence of male central precocious (CPP) is increasing. In this study, we explore CPP identify key clinical auxological indicators for organic (OCPP). Design A retrospective registry-based study. Methods The medical records 43 treated with Helsinki University Hospital between 1985 2014 were reviewed. Clinical, auxological, endocrine data patients included in analyses. Results Based on brain MRI, 26% had OCPP. Between 2010 2014, was 0.34 per 10 000 (95% CI 0.20-0.60). 1990 increased (incidence rate ratio [IRR] 1.10, P = .001). This increase driven by rising idiopathic (ICPP) (IRR 1.11, 95% 1.05-1.19, < .001), while OCPP remained stable (P .41). Compared ICPP, .006), shorter .003), higher basal serum testosterone levels .038). Combining 2 to 4 these readily available cues resulted good excellent (all, area under curve 0.84-0.97, .001) overall performance, differentiating etiology from idiopathic. Conclusions estimated 000, cases associated intracranial pathology. ICPP rates. Patients characterized stature, levels, providing valuable differentiation addition MRI. Abstract

Purpose

Current knowledge of the characteristics puberty beyond age at menarche and thelarche is limited, particularly within population-based cohorts. Secular trends concerns health effects early reinforce value contemporary studies characterizing timing, tempo, duration, synchronicity puberty.

Methods

The Childhood to Adolescence Transition Study a unique Australian cohort individuals followed annually from late childhood adolescence, with up eight assessments pubertal stage 9 19 years (N = 1,183; 636 females). At each assessment, females reported their Tanner Stage breast pubic hair development, while males on genital/pubic development. Nonlinear mixed-effects models characterized trajectories were used derive individual's estimates synchronicity. Parametric survival estimate overall duration

Results

Timing mid-puberty (Tanner 3) ranged 12.5 13.5 years, developing approximately 6 months before males. Pubertal tempo (at mid-puberty) was similar across sex (between half one per year), but slightly shorter in Most exhibited asynchronous changes

Discussion

Estimates timing are consistent reports cohorts two or more decades ago, suggesting stabilization certain predominantly White populations. However, our understanding individual differences (e.g., physical changes) remains limited.

Abstract Purpose To identify cut-off for basal LH levels and pelvic ultrasound uterine ovarian parameters indicating an Hypotalamic–Pituitary–Gonadal (HPG) axis activation as diagnostic of Central Precocious Puberty (CPP). Methods 248 girls referred suspected precocious/early puberty who had undergone a GnRH stimulation test were enrolled divided into three groups: Premature Idiopathic Thelarche (PIT), CPP, Early (EA). For every patient serum Luteinising Hormone (LH) Follicle Stimulating (FSH), LH/FSH ratio ultrasonographic also collected. Through the use Receiver Operating Curves (ROCs) sensitivity (Se) specificity (Sp) LH, FSH, evaluated at each level Area Under Curve (AUC) was measured. Results Basal model ≥0.14 mIU/mL reached highest predictability (90.6% 78.2%, Se Sp, respectively). ≥0.1 showed 85.90% 78.14%, while FSH (≥2.36 mIU/mL) lowest predictability, with less favourable (71%) (70.5%). Cut-off point length 35 mm, (83.5% 42.9% respectively) calculated. volumes, ROC curves very low specificity. Conclusion A single measurement under limit may be adequate to exclude HPG CPP.
