Abstract Conventional, brightfield-microscopic semen analysis provides important baseline information about sperm quality of an individual; however, it falls short identifying subtle subcellular and molecular defects in cohorts “bad,” defective human animal spermatozoa with seemingly normal phenotypes. To bridge this gap, is desirable to increase the precision andrological evaluation humans livestock animals by pursuing advanced biomarker-based imaging methods. This review, spiced up occasional classic movie references but seriously scholastic at same time, focuses mainly on biomarkers altered male germ cell proteostasis resulting post-testicular carryovers proteins associated ubiquitin-proteasome system. Also addressed are redox homeostasis, epididymal maturation, sperm–seminal plasma interactions, surface glycosylation. Zinc ion homeostasis-associated sperm-borne components, including elements neurodegenerative pathways such as Huntington Alzheimer disease, discussed. Such spectrum biomarkers, imaged highly specific vital fluorescent probes, lectins, antibodies, reveals both obvious chromatin, deoxyribonucleic acid, accessory structures head tail. Introduction next-generation image-based flow cytometry into research clinical andrology will soon enable incorporation machine deep learning algorithms end point developing simple, label-free methods for diagnostics high-throughput phenotyping economically animals.

Male infertility accounts for approximately 40% of cases. There are many causes male infertility, including environmental factors, age, lifestyle, infections, varicocele, and cancerous pathologies. Severe oligozoospermia, cryptozoospermia, azoospermia (obstructive non-obstructive) identified as severe factor once considered conditions sterility. Today, in vitro fertilization (IVF) techniques the only treatment strategy cases which new methodologies have been developed manipulation spermatozoa to achieve increase success rates. Forecasted increases in the prevalence and severity of extreme weather events accompanying changes climatic behavior pose potential risk to reproductive capacity humans animals ecological agricultural significance. While several studies have revealed that heat stress induced by challenges such as testicular insulation can elicit a marked negative effect on male system, particularly production spermatozoa, less is known about immediate impact function following subchronic whole-body exposure elevated ambient temperature. To address this knowledge gap, we exposed unrestrained mice conditions emulate wave (daily cycle 8 h at 35 °C followed 16 25 °C) for period 7 d. Neither testes or epididymides heat-exposed exhibited evidence gross histological change, similarly, spermatozoa males retained their functionality ability support embryonic development. This case report presents an illustrative account of a couple experiencing secondary infertility attributed to the fragmentation DNA in sperm. Secondary infertility, inability conceive after having previously successfully conceived child, can be due various factors, including male issues. Sperm (SDF) has emerged as major factor influencing fertility, resulting poor embryo development and lower pregnancy rates. is about use advanced assisted reproductive technologies, specifically physiological intracytoplasmic sperm injection (PICSI) (ICSI), treat caused by DNA. PICSI enables identification selection spermatozoa with optimal functional integrity using hyaluronan, natural binding substance. Preparing dish requires skill precision. exhibiting high index were excluded from process enhance potential. The underwent controlled ovarian stimulation, egg retrieval, ICSI PICSI. treatment resulted successful conception singleton pregnancy. Subsequent prenatal monitoring indicated healthy progression, ultimately culminating delivery baby girl at term. highlights efficacy integrating method preceding ICSI, cases related SDF. Further research larger-scale studies are warranted approve findings this establish broader applicability approach.

Result:The first method showed statistically significant higher motility DGC/su (p<0.001), while other significant, lower fewer abnormalities DGC/Z (p<0.001).We discovered no connection between defective morphology damage. There is link damage, indicating that are more likely be normal DNA. Conclusion:According results current study, use DGC/swim-up procedures improved motility, morphology, integrity samples, which may increase probability a successful pregnancy.
