This paper provides a comprehensive summary of the toxic effects drugs on male reproductive system, with special focus mechanisms testicular and sperm damage caused by chemotherapeutic agents, antibiotics, immunosuppressants. Drug-induced toxicity usually manifests through multiple pathways including direct injury, interference hormone regulation, oxidative stress, DNA damage. These can lead to impaired spermatogenesis, decreased function, long-term infertility, thereby seriously affecting health. Specifically, drugs, such as cisplatin cyclophosphamide, have been widely documented cause testes, leading significant decreases in quantity quality. Antibiotics nonsteroidal anti-inflammatory also negatively affect function mitochondrial inducing stress cells. Although important advances made recent years study drug-induced toxicity, further exploration is needed assess variability individual responses develop protective measures. Future research should developing accurate assessment methods, customized therapeutic regimens, enhanced fertility protection strategies, use antioxidants methods for cryopreservation testes sperm. Interdisciplinary collaboration will provide new solutions balancing disease treatment preservation, especially high-risk anticancer therapies, where achieving both efficacy health be an clinical challenge.
