The neuropeptides arginine vasopressin (AVP) and oxytocin (OXT) are key regulators of social behaviour across vertebrates. However, much our understanding how these neuropeptide systems interact with is centred around laboratory studies which fail to capture the physiological challenges living in wild. To evaluate relationships between wild, we studied groups cichlid fish Neolamprologus pulcher Lake Tanganyika, Africa. We first used SCUBA observe focal group members then measured transcript abundance components AVP OXT different brain regions. While often associated male-typical behaviours, found that dominant females had higher expression avp its receptor (avpr1a2) preoptic area compared either males or subordinates sex. Dominant also generally highest levels leucyl-cystinyl aminopeptidase (lnpep)-which inactivates OXT-throughout brain, potentially indicating greater overall activity (i.e., production, release, turnover) system females. Expression receptors did not differ ranks. visited brood chamber more lower a (oxtra) suggesting negative relationship signalling parental care this species. Overall, results advance complex behaviours neuroendocrine under natural settings.

Abstract Surge release of luteinizing hormone (Lh) from the pituitary is essential for fertility as it triggers ovulation. While secretoneurin (SN) a phylogenetically conserved secretogranin-2-derived peptide that stimulates Lh, its role in ovulation has not been established. To directly compare periovulatory changes classical hormones to emerging reproductive neuropeptides SNa and SNb, simultaneous mass spectrometry measurement 9 peptides 5 steroids was conducted female zebrafish. Regression analysis indicated levels SNa1–34 brain peaked when type 3 gonadotropin-releasing (Gnrh3) increased (R2 = 0.71) at time Lh surge, 3.5 h before Levels naturally occurring derivative SNa1–14 were highest ovulation, while SNb1–31 invariable. The bioactivities investigated. After injection females had isolated males, 61% (11/18) ovulated within 6 h, which like effects analog human chorionic gonadotropin (72%; 13/18 females). induces by increasing time-dependent expression gnrh3 brain, likely direct stimulation alpha (cga) b (lhb) subunit pituitary, via subsequent increase nuclear progesterone receptor (npr) ovaries. Reproduction is a complicated affair, requiring the well-timed coordination of hormone levels, brain signals and behavior. The humble zebrafish makes it look easy – their frequent reproduction abundant offspring contribute to popularity this species in research. Estrogens are among most important steroid hormones involved reproduction. Fish have unique quirk estrogen production system due gene duplication: they two specialized versions estrogen-producing enzyme whereas mammals birds just one. One version primarily found gonads fish, including ovaries, where critical for proper development function. other brain, but its role there not so clear. Could tissue-specific fish impact reproduction? Katherine Shaw (University Ottawa, Canada) colleagues from same institution Sun Yat-Sen University, China, investigated whether disruption can really throw wrench into gears reproduction. To tackle question, genetically modified disrupt that codes brain. This approach allowed them specifically inhibit while leaving ovarian intact. Accordingly, measured lower levels estradiol (the primary fish) these mutated female fish. They also observed were about four times slower start releasing eggs when paired with normal male during morning mating session. Without supplying behavior was thrown off. But result led another question how does drop bring delay egg release? This an duo signaling molecules arginine vasopressin oxytocin join story. These produced pathways control reproductive gamete release. Since release delayed females, researchers suspected and/or might link timing both reduced. Was between reduced eggs? perhaps restore by increasing or fish's body?To find out, team injected (with estradiol) either mating. vasopressin-injected now released within more typical timeframe. restoration showed key signal linking When fluorescently labeled different cell types; however, cells close proximity, potentially allowing brain-derived estrogens stimulate nearby vasopressin. What we learned all this? encodes brain-specific production, which is, help vasopressin, prompt has laid foundation further explore roles as well fishes, spawn new ideas sex steroids influence
