Background Psoriasis is commonly associated with metabolic dysfunction-associated steatotic liver disease, raising concerns about the hepatic effects of systemic treatments on psoriasis and its comorbid conditions. This study evaluates stiffness measurement (LSM) alterations identifies predictors abnormal LSM in psoriatic patients following treatments, including biologics methotrexate. Methods prospective cohort based PSOWCH database (Psoriasis Cohort West China Hospital). We initially included who had undergone sound touch elastography (STE), then recruited STE before treatment reassessed them after at least six months. Three subgroups were formed (interleukin inhibitors, tumor necrosis factor methotrexate), classifying post-treatment outcomes using threshold values 6.5 kPa 10.3 kPa. Results Among 52 patients, overall significantly increased during follow-up. Univariate regression analysis showed that age, gender, severity, arthritis status, current type not correlated cutoff In multivariate model, body mass index (BMI) was identified as a risk for ≥ (odds ratio [OR], 1.26; 95% CI, 1.04 to 1.60, P=0.031). Metabolic dysfunction-associated fatty liver disease (MAFLD) is the most common cause of chronic worldwide, with a multifactorial etiology. This study aims to evaluate associations between various sociodemographic variables, healthy habits, and stress risk scale values for MAFLD. A descriptive, cross-sectional was conducted on 16,708 Spanish workers assess how variables (age, gender, socioeconomic status), habits (smoking, Mediterranean diet adherence, physical activity), correlate from three MAFLD scales: index (FLI), hepatic steatosis (HSI), lipid accumulation product (LAP). Aim: While cardiometabolic disorders and metabolic dysfunction-associated steatotic liver disease (MASLD) frequently coexist, the genetic connections causes are not clearly understood. This study aimed to explore their shared architecture elucidate mechanisms driving comorbidity. Methods: Using summary statistics from genome-wide association studies (GWASs) on MASLD 29 traits (CMTs), we assessed correlation causality, identified loci, genes, pathways, cell types, tissues. Additionally, biological were uncovered using single-cell RNA sequencing data. Results: Significant correlations detected between 17 CMTs, encompassing diseases, glucose, lipids, adiposity, inflammatory markers, after adjusting for multiple testing (p .adjust < 0.05). Cross-trait analysis yielded a total of 166 risk SNPs (including those located in or near TRIB1 , LPL PNPLA3 GCKR PPARG ). Subsequent colocalization highlighted 73 loci associated with both rs429358 (APOE ) consistently prioritized HyPrColoc. Common genes (such as NPC1 MST1R TMBIM1 IRAK1BP1 L3MBTL3 RBM6 RGS19 ), significant enrichment cholesterol metabolism, glucose immune inflammation, long-term depression.
