Extreme morphological disparity within Mollusca has long confounded efforts to reconstruct a stable backbone phylogeny for the phylum. Familiar molluscan groups—gastropods, bivalves, and cephalopods—each represent diverse radiation with myriad morphological, ecological, behavioral adaptations. The phylum further encompasses many more unfamiliar experiments in animal body-plan evolution. In this work, we reconstructed living on basis of metazoan BUSCO (Benchmarking Universal Single-Copy Orthologs) genes extracted from 77 (13 new) genomes, including multiple members all eight classes two high-quality genome assemblies monoplacophorans. Our analyses confirm proposed morphology show widespread genomic variation. flexibility likely explains both historic challenges their genomes evolutionary success.

Abstract Mollusca is the second most species-rich animal phylum, but pathways of early molluscan evolution have long been controversial 1–5 . Modern faunas retain only a fraction past forms in this hyperdiverse and long-lived group. Recent analyses 6–8 consistently recovered fundamental split into two sister clades, Conchifera (including gastropods, bivalves cephalopods) Aculifera 9 , comprising Polyplacophora (‘chitons’) Aplacophora. Molluscan toto characterized by plasticity body-plan characters 10 historically aculiferans interpreted as more conservative 10,11 The few completely preserved aculiferan or aculiferan-like fossils from Palaeozoic 12–19 largely regarded transitional that inform questions character polarity between extant polyplacophoran aplacophoran body 20,21 history aculiferans, morphological ecological range they occupied, remain inadequately sampled. Abstract Sperm structure among species in each Class of Aplacophora is highly conserved but between the two Classes radically different. This together with numerous morphological differences groups, suggests a long separation and likely paraphyletic relationship, which has been suggested past not supported by molecular analyses. All Caudofoveata examined have unique externally-fertilizing ect-aquasperm found nowhere else Animal Kingdom. Solenogastres fertilize internally introsperm like those described for Epimenia australis differ details length number specific components, providing insights to relationships them. Furthermore, solenogaster shares at least nine characters bilaterian lineage Nemertodermatida, none these sperm Caudofoveata, Polyplacophora (chitons), or Xenoturbellida few Acoela.
