Background: Skin cutaneous melanoma (SKCM) is one of the most common malignancies, which incidence increasing. Cuproptosis a new type programming cell death recently reported, may affect progression SKCM. Method: The mRNA expression data were obtained from Gene Expression Omnibus and Cancer Genome Atlas databases. We constructed prognostic model according to cuproptosis-related differential genes in Finally, real-time quantitative PCR was performed verify patients with different stages melanoma. Results: detected 767 based on 19 genes, screened out 7 construct model, including three high-risk (SNAI2, RAP1GAP, BCHE), four low-risk (JSRP1, HAPLN3, HHEX, ERAP2). Kaplan-Meier analysis indicated that SKCM signals had better prognosis. Encyclopedia Genomes results manifested are not only involved T receptor signaling channel, natural killer mediated cytotoxicity, but also chemokine pathway B pathway. In our risk scoring receiver operating characteristic (ROC) values three-time nodes 0.669 (1-year), (3-year) 0.685 (5-year), respectively. Moreover, tumor burden mutational immunology function, stemness characteristics drug sensitivity have significant differences between group group. level SNAI2, RAP1GAP BCHE stage Ⅲ+Ⅳ significantly higher than Ⅰ+Ⅱ patients, while JSRP1, HHEX ERAP2 more remarkable patients. Conclusion: summary, we suggest cuproptosis can regulate immune microenvironment prognosis offer basic theory for survival studies clinical decision-making potentially therapeutic drugs.

Coronary artery disease (CAD) usually coexists with subclinical cerebrovascular diseases given the systematic nature of atherosclerosis. In this study, our objective was to predict progression white matter hyperintensity (WMH) and find its risk factors in CAD patients using coronary calcium (CAC) score. We also investigated relationship between CAC score WMH volume different brain regions. We evaluated 137 who underwent computed tomography angiography (CCTA) two magnetic resonance imaging (MRI) scans from March 2018 February 2023. Patients were categorized into progressive (n = 66) nonprogressive groups 71) by change first second MRI. collected demographic, clinical, data for analysis. Independent identified logistic regression. Three models predicting developed assessed. Finally, divided based on their total (0 <100, 100 400, > 400) compare changes nine regions.Alcohol abuse, maximum pericoronary fat attenuation index (pFAI), CT-fractional flow reserve (CT-FFR), grade independently predicted (p < 0.05). The regression model all four variables performed best (training: AUC 0.878, 95% CI: 0.790, 0.938; validation: 0.845, 0.734, 0.953). An increased came significantly higher brain, corpus callosum, frontal, parietal occipital lobes 0.05).This study demonstrated application CCTA-derived elderly people (≥60 years) CAD.

Abstract Cuproptosis, a newly defined cell death process, represents novel modality with significant therapeutic potential in cancer treatment. Nevertheless, the modest concentration and transient half‐life of copper ions bloodstream constrain their efficient delivery into tumor cells. In this study, copper‐based prussian blue nanostructure loaded serine metabolic inhibitor (NCT‐503@Cu‐HMPB) is constructed for selectively inducing cuproptosis combined disrupting metabolism. Released within cells, NCT‐503 found to inhibit cellular metabolism GSH production, ultimately causing dysfunction, redox imbalance, increased formation Cu + that disrupts mitochondrial respiration chain, lipoylated protein dihydrolipoamide S‐acetyltransferase (DLAT) aggregation consequential iron‐sulfur cluster loss, which leads proteotoxic stress results death. The findings provide paradigm therapy based on reprogramming, offering prospects development innovative nanotherapeutic platforms future.

Inflammation is one of the most important characteristics tumor tissue. Signatures based on inflammatory response-related genes (IRGs) can predict prognosis and treatment response in a variety tumors. However, clear function IRGs triple negative breast cancer (TNBC) still needs to be explored.IRGs clusters were discovered via consensus clustering, prognostic differentially expressed (DEGs) across utilized develop signature using least absolute shrinkage selection operator (LASSO). Verification analyses conducted show robustness signature. The expression risk was identified by RT-qPCR. Lastly, we formulated nomogram improve clinical efficacy our predictive tool. The signature, comprised four genes, developed shown highly correlated with prognoses TNBC patients. In contrast performance other individual predictors, that remarkably superior. Also, ImmuneScores elevated low-risk group. immune cell infiltration showed significant difference between two groups, as did checkpoints. The could act biomarker provide momentous reference for therapy TNBC.
