ABSTRACT Microglia are the primary resident immune cells of central nervous system (CNS) that respond to injury and infections. Being critical CNS homeostasis, microglia also have been shown contribute neurodegenerative diseases brain cancer. Hence, regarded as a potential therapeutic target in diseases, resulting an increased demand for reliable vitro models. Two human cell lines (HMC3 C20) being used multiple studies, however, knowledge their biological immunological characteristics remains limited. Our aim was identify compare changes these immortalised under normal physiological immunologically challenged conditions. Using high‐resolution quantitative mass spectrometry, we examined in‐depth proteomic profiles non‐stimulated LPS or IFN‐γ HMC3 C20 cells. findings reveal responded both treatments through upregulation immune, metabolic, antiviral pathways, while showed response associated with mitochondrial activities. Additionally, secretome analysis demonstrated release IL‐6 LPS, treatment resulted altered kynurenine pathway activity, highlighting distinct metabolic adaptations.

The clutch is defined as consecutive days of oviposition. Clutch length an index that reflects ovulation persistence, and highly correlated with egg production in birds. To identify the genetic markers associated geese, two experiments were conducted. In first experiment, 200 Zi geese selected, all 230 old, selected from same batch raised individually environment. Data egg-laying traits recorded. After laying period, three longest lengths to form group (LCG) shortest formed short (SCG). second ovaries six collected for transcriptomic metabolomic analyses. results showed large (LCL) average (ACL) positively number (EN) (P < 0.01; r = 0.63 0.60, respectively). Large (LCN) was significantly peak (PEN) (r 0.58, P 0.01) EN 0.01). EN, LCN, LCL, ACL significant differences between groups. Transcriptomic analysis identified 424 differentially expressed genes (DEGs). Functional enrichment revealed these DEGs mainly involved neuroactive ligand-receptor interactions, ovarian steroidogenesis, calcium signaling pathways. Further, AVPR1A, FGF14, LHCGR predicted key regulating LCL. Metabolomic 349 differential metabolites (DMs) both positive negative ion modes. Pyruvate, isocitric acid, D/L‑serine, 3-phospho-d-glycerate, succinate, glycine, glutamic acid enriched pathways TCA cycle. Integration data critical gene-metabolite pairs, including ACSL4-phosphoenolpyruvate, implicated LCL regulation. summary, this study provides new insights into molecular affecting geese.

Abstract Cancer is one of the leading causes mortality worldwide, highlighting urgent need for a deeper molecular understanding disease's heterogeneity and development personalized treatments. Since its establishment in 2017, Human Pathology Atlas has been instrumental linking gene expression profiling with patient survival outcomes, providing system-level insights experimental validation across wide range cancer research. In this updated analysis, we analysed profiles 6,918 patients 21 types using latest annotations. Our refined approach enabled us to offer an list prognostic genes human cancers, focus on hepatocellular, renal colorectal cancers. To strengthen reliability our findings, integrated data from 10 independent cohorts, creating cross-validated, reliable collection genes. By applying systems biology approach, identified that outcomes kidney clear cell carcinoma (KIRC) liver hepatocellular (LIHC) are strongly associated profiles. We also developed regulatory network specifically KIRC LIHC enhance utility The version lays foundation precision oncology treatment strategies.
