The economic importance of lumpfish ( Cyclopterus lumpus ) is increasing, but several aspects its immune responses are not well understood. To discover genes and mechanisms involved in the antiviral response, fish were intraperitoneally injected with either viral mimic polyinosinic:polycytidylic acid [poly(I:C)] or phosphate-buffered saline (PBS; vehicle control), head kidneys sampled 24 hours post-injection (hpi) for transcriptomic analyses. RNA sequencing (RNA-Seq) (adjusted p-value <0.05) identified 4,499 upregulated 3,952 downregulated transcripts poly(I:C)-injected compared to PBS-injected fish. Eighteen as differentially expressed by RNA-Seq included a qPCR study that confirmed upregulation encoding proteins response functions (e.g., rsad2 downregulation jarid2b potential cellular process functions. In addition, transcript expression levels 12 members interferon regulatory factor (IRF) family [seven which poly(I:C)-responsive this study] analyzed using qPCR. The Janus kinase/signal transducers and activators of transcription (JAK-STAT) system regulates several biological processes by affecting genes as a response to cytokines growth factors. In the present study, we have characterized STAT in lumpfish (Cyclopterus lumpus L.), belonging order Perciformes, investigated regulation JAK-STAT signaling pathway upon exposure bacteria (Vibrio anguillarum) poly(I:C), latter mimicking antiviral responses.Characterization evolutionary analyses STATs were performed phylogeny, protein domain, homology similarity synteny analyses. Antibacterial responses performing KEGG analysis. We observed that stat1a, 2, 3, 4, 5a, 5b, 6. Transcriptome-wide showed most components lumpfish. il-6, il-10, il-21, iκBα stat3 upregulated 6 hours post (hpe) against while type I interferons (IFNs), irf1, irf3, irf10, stat1 2 24 hpe poly(I:C).Our findings shed light on diversity evolution data show are highly conserved among fish, including transcriptome-wide lay groundwork for future research into functional significance these regulating critical make an important basis development prophylactic measure such vaccination, which is needed since it vulnerable both bacterial viral diseases.
