The production of viable nucleopolyhedrovirus in the feces infected lepidopteran larvae represents a poorly understood route for virus transmission prior to host death. In present study, we examined presence fourth-instar Spodoptera frugiperda with Nicaraguan isolate multiple (SfMNPV-NIC). Feces increased samples taken at 2 6 days post-inoculation but was significantly lower insects compared controls. Second instars experienced 3.9 68.3% polyhedrosis disease following consumption collected 2–5 post-inoculation, which subsequently fell 29.1% 6-day sample. Calibration insect bioassay using OB-spiked indicated that concentration OBs varied between 5.4 × 102 and 4.4 105 OBs/100 mg fourth instars. Quantitative PCR analysis fecal 0 7629 copies/mg amplification targeted polyhedrin gene. However, no correlation detected qPCR estimates time sample collection or quantity collected. were positively correlated prevalence lethal infection observed bioassay, weak several did not amplify. assay provided 5- 10-fold than indicating inhibition reaction loss material during processing. greenhouse experiment, 2.5–48.3% second-instar acquired 24 h period feeding on maize plants instar had deposited their 3 4 post-infection, respectively. These findings highlight potential OB-contaminated as source biologically significant quantities inoculum death represent an additional contribution biological control pests by these pathogens.

The silkworm, Bombyx mori , is a crucial model insect in agriculture and biological research. Tetraspanins, known for their effects regulating cellular functions like cell signalling, adhesion, migration diffusion, take on role viral dynamics, influencing both spread entry into host cells. In this study, tetraspanin gene called BmTsp.C from the silkworm genome was identified investigated. Tissue profiles showed that has highest transcription level midgut, with marked increase following infection. immunofluorescence localization suggested primarily distributed membrane. Additionally, overexpression of BmN cells facilitated proliferation BmNPV. Meanwhile, siRNA-mediated knockdown could inhibit proliferation. addition, BmTsp .C at individual further validated remarkable effect during infestation. Furthermore, regulate expression apoptosis-related genes. Results flow cytometry indicated decrease number apoptotic after . Taken together, our results demonstrated as an important factor Tetraspanin-enriched microdomains, exerts significant influence BmNPV, most likely through apoptosis pathway.
