Background It has been reported that COVID-19 patients have an increased risk of developing IBS; however, the underlying genetic mechanisms these associations remain largely unknown. The aim this study was to investigate potential shared SNPs, genes, proteins, and biological pathways between IBS by assessing pairwise correlations cross-trait analysis. Materials methods We assessed correlation three phenotypes using linkage disequilibrium score regression (LDSC) high-definition likelihood (HDL) methods. Two different sources data were combined METAL, Multi-trait analysis GWAS (MTAG) method applied for multi-trait enhance statistical robustness discover new associations. Independent loci examined genome-wide complex trait (GCTA)-conditional joint (COJO), multi-marker genomic annotation (MAGMA), functional mapping (FUMA), integrating various QTL information further identify genes proteins. Gene set variation (GSVA) employed compute pleiotropic gene scores, with immune infiltration algorithms, categorized into high low groups. Results found a positive infection, hospitalization, IBS. Subsequent identified nine significantly associated loci. Among these, eight variants closely related comorbidity COVID-19. also highlighted four 231 proteins susceptibility through analytical strategies stratification approach populations.
