Ferroptosis is a novel form of cell death characterized by unlimited accumulation iron-dependent lipid peroxides. It often accompanied disease, and the relationship between ferroptosis immune cells regulation has been attracting increasing attention. Initially, it was found in cancer research that inhibition regulatory T (Treg) promotion CD8+ jointly promoted formation an immune-tolerant environment tumors. T-cell subsequently to have immunoregulatory effects other diseases. As autoimmune disease imbalance, attracted attention for its potential regulating balance lupus nephritis. This article reviews metabolic processes within different subsets nephritis (LN), including follicular helper (TFH) cells, (Th)17 Th1 Th2 Treg reveals these cellular metabolisms not only facilitate imbalance but are also closely associated with occurrence ferroptosis. Consequently, we hypothesize targeting pathways could become direction effectively treating altering differentiation incidence

Rheumatoid arthritis (RA) is a widely prevalent rheumatic condition causing joint inflammation and damage. RNA methylation plays an important role in RA. Herein, we intended to investigate the function of methyltransferase-like 3 (METTL3) its N6-methyladenosine (m6A) regulation for ras-related C3 botulinum toxin substrate 2 (RAC2) MH7A cells were treated with TNF-α establish RA cell model. The expression analysis was performed by RT-qPCR western blot. Cellular behaviors examined CCK-8 assay, flow cytometry, wound healing assay transwell assay. Oxidative stress assessed detecting associated indicators. Inflammatory cytokines measured via enzyme-linked immunosorbent (ELISA). Interaction between METTL3 RAC2 analyzed immunoprecipitation (RIP) MeRIP highly expressed tissues TNF-α-stimulated cells. Knockdown enhanced apoptosis reduced proliferation, migration, invasion after treatment. downregulation suppressed oxidative inflammatory response TNF-α-treated promoted through m6A methylated modification, METTL3/RAC2 could activate AKT pathway. overexpression reversed effects knockdown on motility, inflammation. above results demonstrated that facilitated progression downregulating dependent mechanism

Rheumatoid arthritis (RA) is a chronic disease of widespread concern worldwide, and there an urgent need to develop sensitive methods for the rapid detection RA. Previous studies have shown that RA closely related lysosomal dysfunction. Lysosomal viscosity important microenvironmental parameter reflecting state lysosomes, but due lack probes demonstrate correlation between RA, changes in during remain unclear. For this purpose, we report herein lysosome-targeted near-infrared fluorescent molecular rotor probe DSMP investigate This utilizes dicyanomethylene-4H-benzothiopyran as electron acceptor fluorophore piperazine unit donor targeting group lysosomes. In addition, shows strong solid fluorescence response can effectively target lysosomes detect live cells. Based on this, established mouse model using λ-carrageenan. Mice imaging show quickly image tissues exhibit signals significantly brighter than those normal joint tissues. indicates increase RA; therefore, serve indicator be effective tool research.
