This study investigated seasonal variations of thraustochytrids in rhizosphere soil collected from two mangrove species Rhizophora apiculata and Avicennia marina natural planted stands. Thraustochytrid counts were higher the stand than site. The varied seasonally, being highest post-monsoon season followed by summer, monsoon, pre-monsoon. exhibited positive correlations (p < 0.01) with Total heterotrophic bacteria (THB), azotobacters, actinobacteria, fungi, yeasts, Trichoderma. However, a negative correlation was observed cyanobacteria. also had silt, clay, nitrogen, phosphorus, potassium, chromium, copper, magnesium, cadmium, zinc redox potential, but, temperature, pH, pore water salinity, total organic carbon sand content samples. In present study, 113 thraustochytrid strains isolated habitats. only 48 pure cultures survived after sub-cultured three times. Based on survivability, color, shape, isolates each sampling site selected. predominant 24 identified based their morphological, molecular characteristics classified under five genera: Thraustochytrium, Schizochytrium, Botryochytrium, Parietichytrium, Aurantiochytrium. Among isolates, Aurantiochytrium sp. (AKTSK-06) produced biomass 15.71 g/L (January-March, 2023), (PVTSK-03) accumulated lipid 61.33%. Thraustochytrids found to contain Omega-3 poly unsaturated fatty acids (PUFAs), such as EPA up 8.89% (VRTSK-01), DPA 9.65% (AKTSK-03), DHA 47.46% (AKTSK-06). Thus, mangroves provide an ideal ecological niche for abundant supply omega-3 potential industrial applications.

ABSTRACT Thraustochytrids, diverse marine unicellular protists encompassing over 10 recognised genera, are renowned for synthesising polyunsaturated fatty acids (PUFAs), with content and composition varying substantially across genera. While PUFAs known to be produced via PUFA synthase (PUFA‐S) and/or elongase/desaturase (ELO/DES) pathways, the distinctions in genes involved remain unexplored. This study analysed biosynthetic 19 thraustochytrid strains six categorising them into four types. Type I exclusively utilises ELO/DES pathway, II employs both PUFA‐S complete while Types III IV primarily rely on PUFA‐S, lacking canonical Δ9 desaturase missing most elongase enzymes. Notably, ATP‐citrate lyase ( ACLY ) exclusive II, β‐carotene hydroxylase CrtZ is absent these absence suggests alternative acetyl‐CoA supply pathways IV, whereas implies either a lack of specific xanthophylls or II. Synteny analysis revealed conserved genomic organisation genes, indicating shared evolutionary trajectory. provides insights genetic diversity underlying biosynthesis thraustochytrids, proposing putative lineages.
